摘 要:钙离子(Ca^(2+))是参与细胞信号转导重要的第二信使,Ca^(2+)失调可能会导致病理变化。钙池操纵Ca^(2+)内流(SOCE)是一种胞外-胞内信号转导的独特方式,可以调控细胞内外Ca^(2+)的动态平衡,异常的钙信号与恶性肿瘤细胞上皮-间充质转化、肿瘤血管生成、侵袭迁移及肿瘤免疫密切相关。钙释放激活钙通道调节分子1(Orai1)是一种参与SOCE的Ca^(2+)通道,在SOCE激活过程中Orai1主要受基质相互作用分子(Stim)蛋白尤其是Stim1的调控,在几种细胞类型的Ca^(2+)稳态中起关键作用。Stim1异常表达不仅可导致包括恶性肿瘤在内的多种疾病发生,还与恶性肿瘤发生耐药的机制息息相关,有望为肿瘤的临床治疗提供新靶点。Ca^(2+)is a crucial second messenger involved in cellular signal transduction,and its dysregulation can lead to various pathological conditions.Store-operated Ca^(2+)entry(SOCE)represents a unique mechanism of extracellular-intracellular signaling that regulates the dynamic balance of Ca^(2+)between the intracellular and extracellular environments.Aberrant calcium signaling is closely linked to processes such as epithelial-mesenchymal transition in malignant tumor cells,tumor angiogenesis,invasion,metastasis,and tumor immunity.Calcium release-activated calcium channel protein 1(Orai1),a Ca^(2+)channel involved in SOCE,plays a pivotal role in maintaining Ca^(2+)homeostasis in various cell types,primarily regulated by stromal interaction molecule(Stim),especially Stim1.Abnormal expression of Orai1 not only leads to the occurrence of various diseases,including malignant tumors,but is also closely related to the mechanism of drug resistance in malignant tumors,which is expected to provide new targets for the clinical treatment of tumors.
