摘 要:目的探究加味柴胡当归汤对大鼠肝纤维化的治疗效果及其机制。方法从52只SD大鼠中随机选取10只作为空白组,其余大鼠腹腔注射40%(体积分数)CCl4橄榄油构建肝纤维化(HF)模型,验证造模成功后,剩余大鼠随机分为模型组和加味柴胡当归汤低、中、高剂量组,给予不同剂量汤剂进行治疗。实验过程中观察记录大鼠外观表态,治疗结束后,处死大鼠取材。收集血清检测大鼠肝功能和透明质酸酶(HA)、层粘连蛋白(LN)、Ⅲ型前胶原肽(PⅢP)和Ⅳ型胶原(CⅣ)水平;采用ELISA检测血清和肝组织中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、α平滑肌肌动蛋白(α-SMA)、Ⅰ型胶原(COL-Ⅰ)水平;利用Western Blot和qRT-PCR实验测定肝组织中PI3K、AKT、Bcl-2蛋白和mRNA的表达。结果实验结束后,模型组大鼠血清中谷丙转氨酶(GPT)、谷草转氨酶(GOT)水平较空白组升高,白蛋白(ALB)水平下降(P<0.05);与模型组相比,加味柴胡当归汤低、中、高剂量组大鼠血清中GPT、GOT水平下降,而ALB水平上升(P<0.05),其中高剂量组与空白组差异无统计学意义(P>0.05)。与空白组相比,模型组大鼠血清中HA、LN、PⅢP和CⅣ水平升高(P<0.05);加味柴胡当归汤低、中、高剂量组HA、LN、PⅢP和CⅣ水平相比于模型组降低(P<0.05),且高剂量组与空白组差异无统计学意义(P>0.05)。模型组血清中TNF-α、IL-6、IL-1β和肝组织中α-SMA、COL-Ⅰ表达水平相比于空白组上升(P<0.05),加味柴胡当归汤低、中、高剂量组较模型组血清TNF-α、IL-6、IL-1β及肝组织α-SMA、COL-Ⅰ表达均下降(P<0.05),高剂量组与空白组差异无统计学意义(P>0.05)。模型组大鼠肝组织中PI3K、AKT、Bcl-2的蛋白及mRNA水平相较于空白组均升高(P<0.05),加味柴胡当归汤低、中、高剂量组大鼠肝组织中PI3K、AKT、Bcl-2蛋白及mRNA水平对比模型组均下调(P<0.05),且高剂量组与空白组差异无�Objective To investigate the effect and mechanism of modified Chaihu Danggui Decoction in treating hepatic fibrosis(HF)rats.Methods Ten rats were randomly selected from 52 SD rats as the blank group,and the remaining rats were intraperitoneally injected with 40%CCl 4 olive oil to construct HF models.After successful modeling,the remaining rats were randomly divided into the model group,the low-dose,medium-dose,and high-dose modified Chaihu Danggui Decoction groups.Rats’appearance was observed during the experiment,and the rats were sacrificed after treatment for sampling.Liver function and hyaluronidase(HA),laminin(LN),procollagenⅢpeptide(PⅢP)and typeⅣcollagen(CⅣ)of HF in the rats were detected.The levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β),alpha-smooth muscle actin(α-SMA)and typeⅠcollagen(COL-Ⅰ)in serum and liver tissues were measured by ELISA.The protein and mRNA expression of PI3K,AKT and Bcl-2 in liver tissues was determined by Western Blot and qRT-PCR.Results After the end of experiment,serum levels of glutamic-pyruvic transaminase(GPT)and glutamic-oxaloacetic transaminase(GOT)in the model group were higher than those in the blank group,and serum albumin(ALB)level was lower than that in the blank group(P<0.05).Compared with the model group,serum levels of GPT and GOT were lower,and ALB level was higher in the three modified Chaihu Danggui Decoction groups(P<0.05).The difference between the high-dose group and the blank group was not statistically significant(P>0.05).Compared with the blank group,the levels of HA,LN,PⅢP and CⅣwere higher in the model group(P<0.05).Compared with the model group,the four indicators of HF were lower in the three modified Chaihu Danggui Decoction groups(P<0.05),but there was no statistical difference between the high-dose group and the blank group(P>0.05).The levels of TNF-α,IL-6 and IL-1βin serum,the expression levels of SMA and COL-Ⅰin liver tissues of the model group were higher than those in the blank group(P<
