摘 要:试验旨在探究补喂不同水平褪黑素(Melatonin,MT)对绵羊抗氧化能力、血浆中褪黑素含量及尿液中6-羟基褪黑素含量的影响。选择3月龄健康哈萨克公羊25只,随机分为5组,分别为对照组、试验Ⅰ组、试验Ⅱ组、试验Ⅲ组、试验Ⅳ组,每组5只,分别对应补饲0、1、2、4和8 mg/d的褪黑素,为期19 d。在试验第13~16天采集试羊的尿样,第18天上午饲喂前0 h,饲喂后1、2、4 h通过颈静脉采集血样。结果显示,补喂褪黑素2 h后,试验Ⅳ组血浆中总抗氧化力(T-AOC)与试验Ⅰ组相比显著提高(P<0.05);试验Ⅲ、Ⅳ组过氧化氢酶(CAT)活性、超氧化物歧化酶(SOD)活性均显著高于对照组(P<0.05),血浆中丙二醛(MDA)含量显著低于对照组(P<0.05);试验Ⅳ组血浆中谷胱甘肽过氧化物酶(GSH-PX)活性显著高于对照组(P<0.05)。试验羊补喂褪黑素1 h时,对照组及试验组血浆中褪黑素含量均达到峰值,4 h时各试验组血浆中褪黑素含量均显著高于对照组(P<0.05);试验Ⅱ、Ⅲ、Ⅳ组尿液中6-羟基褪黑素排出量与对照组相比均显著提高(P<0.05)。综上所述,补喂褪黑素能够提高绵羊抗氧化能力,血浆和尿液中褪黑素代谢物含量也随着补喂量的增加而增加,综合比较,补喂量以4 mg/d剂量最佳。This experiment was conducted to investigate the effects of different levels of melatonin(MT)supplementation on antioxidant capacity,plasma melatonin content and urine 6-hydroxymelatonin content in sheep.A total of 25 three-month-old healthy Kazakh rams were randomly divided into control group,trial groupsⅠ,Ⅱ,ⅢandⅣ,with 5 lambs in each group.The corresponding diets were supplemented with 0,1,2,4 and 8 mg/d melatonin for 19 days,respectively.Urine samples were collected from experimental sheep on days 13-16,and blood samples were collected through the jugular vein on the morning of day 18,0 hour before feeding,and 1,2,and 4 hours after feeding.The results showed as follows:after 2 h supplementation with melatonin,the plasma total antioxidant capacity(T-AOC)in trial groupⅣwas significantly increased than that in trial groupⅠ(P<0.05).The catalase(CAT)activity and superoxide dismutase(SOD)activity of trial groupsⅢandⅣwere significantly higher than those of control group(P<0.05),and the plasma malondialdehyde(MDA)content was significantly lower than that of control group(P<0.05).The glutathione peroxidase(GSH-PX)activity of trial groupⅣwere significantly higher than that of control group(P<0.05).Plasma melatonin content reached the peak value in control group and experimental groups at 1 h,and plasma melatonin content in experimental groups was significantly higher than that in control group at 4 h(P<0.05).The excretions of 6-hydroxymelatonin in urine of trial groupsⅡ,ⅢandⅣwere significantly increased compared with control group(P<0.05).In summary,supplementation of melatonin can improve the antioxidant capacity of sheep,and the content of melatonin metabolites in plasma and urine also increases with the increase of supplemental feeding,and the optimal dose of supplemental feeding is 4 mg/d.
