摘 要:目的:优化灵芝孢子粉药渣中的多糖提取工艺并研究体外抗氧化活性。方法:采用Box-Behnken试验设计,深入探讨液固比、提取时间及提取温度三个关键参数对多糖提取率的具体作用,确定最佳的提取条件,以优化多糖的提取效率;并对灵芝孢子粉药渣多糖进行体外抗氧化活性研究。结果:灵芝孢子粉药渣多糖的最佳提取工艺为:超声波功率300 W、液固比为120∶1(mL∶g)、提取时间30 min、提取温度65℃。在此提取条件下灵芝孢子粉药渣中多糖提取率达到了6.31%;过氧化氢诱导建立氧化应激细胞模型的实验表明,有一定的抗氧化能力。结论:在灵芝孢子粉药渣的提取过程中,采用超声提取法能有效且充分地释放其中的多糖成分。这些多糖在实验中提升了谷胱甘肽过氧化物酶、超氧化物歧化酶以及过氧化氢酶的活性。此外,它们还积极促进了谷胱甘肽的含量增长,进一步证实了其生物活性的多样性。尤为重要的是,多糖的提取和应用在降低丙二醛含量方面表现出色,为相关领域的研究和应用提供了新的可能性,从而展现了其独特的生物学效应和广泛的应用前景。Object:Optimization of extraction process of polysaccharides from Ganoderma lucidum spore powder dregs.Methods:Box-Behnken test design explores the effect of liquid-solid ratio,extraction time and extraction temperature on the extraction rate of polysaccharide.Determine the optimal extraction conditions to optimize the extraction efficiency of polysaccharides,and study the antioxidant activity of the residue polysaccharide in vitro.Results:In the study of polysaccharide extraction process in the residue of Ganoderma lucidum spore powder,we determined the ultrasonic power of 300 W,liquid-solid ratio of 120∶1(mL∶g),extraction time of 30 min and extraction temperature of 65℃.Under this extraction condition,the polysaccharide extraction rate of Ganoderma lucidum spore powder residue reached 6.31%;the cell model of oxidative stress showed that it has some antioxidant capacity.Conclusions:In the extraction process of ganoderma spore powder residue,the ultrasonic extraction method can effectively and fully release the polysaccharide components.These polysaccharides significantly increased the activity of glutathione peroxidase,superoxide dismutase,and catalase.In addition,they have also actively promoted the growth of glutathione content,further confirming the diversity of their biological activities.More importantly,the extraction and application of polysaccharides performs well in reducing the content of malondialdehyde,providing new possibilities for research and application in related fields,thus demonstrating its unique biological effects and wide application prospects.
