云南部分地区羊十二指肠贾第虫感染情况和分子特征分析

摘  要:为了解云南省绵羊和黑山羊十二指肠贾第虫(Giardia duodenalis)的流行现状和分子特征,评估其人兽共患风险,对云南省楚雄州武定县、红河州个旧市和昭通市永善县采集的156份羊粪便样本(黑山羊55份,绵羊101份),基于gdh基因和bg基因进行套式PCR,经Blast比对确定基因型,使用MrBayes 3.1软件通过贝叶斯推理法(Bayesian inference),采用GTR替换模型(General time reversible,普通时间可逆)构建系统进化树,以Figtree1.4.4软件查看并编辑进化树。结果发现,共检出60份阳性样本(总阳性率为38.46%),其中武定、个旧和永善的阳性率分别为37.50%(9/24)、35.85%(19/53)、40.50%(32/79),不同地区十二指肠贾第虫感染阳性率差异不显著(P>0.05)。黑山羊、绵羊的阳性率分别是40.00%(22/55)、37.62%(38/101),不同品种羊十二指肠贾第虫感染阳性率差异不显著(P>0.05)。经过序列分析,60份阳性样品中有2份为人兽共患A1亚集聚体,58份为集聚体E的不同亚型,E34为优势亚集聚体,其中有1份是E型和A1型的混合感染。结果表明,云南省羊十二指肠贾第虫阳性率略高于国内多数地区,不同地区和不同品种的羊群感染率差异不显著,云南地区存在十二指肠贾第虫人兽共患风险。To explore the prevalence and molecular characteristics of Giardia duodenalis in sheep and black goats in Yunnan province,and to assess the risk of zoonotic diseases,156 faecal samples(55 from black goats and 101 from sheep)were collected from Wuding county in Chuxiong prefecture,Gejiu county in Honghe prefecture,and Yongshan county in Zhaotong city,Yunnan province.Nested PCR was performed based on gdh gene and bg gene.Genotype was determined by comparising of sequenced positive PCR products with those of Giardia duodenalis through Blast.The phylogenetic trees were constructed,with GTR replacement model(General Time Reversible)adopted through Bayesian inference using MrBayes 3.1.The constructed phylogenetic tree were examined and editted with Figtree1.4.4.The results showed that 60 positive samples were detected(total positive rate was 38.46%),among which the positive rates of Wuding,Gejiu and Yongshan were 37.50%(9/24),35.85%(19/53)and 40.50%(32/79),respectively.There was no significant difference in the positive rate of Giardia duodenalis infection among different regions(P>0.05).The positive rates of Giardia duodenalis infection were 40.00%(22/55)in black goats and 37.62%(38/101)in sheep,respectively.There was no significant difference in positive rates between sheep and goats(P>0.05).Sequence analysis showed that among the 60 positive samples,two zoonotic A1 subclusters,whereas 58 various subtypes of E,with E34 being the dominant subcluster,among which one sample was a mixed infection of E and A1.The results showed that the positive rate of Giardia duodenalis in Yunnan province was slightly higher than that in most regions of China,and the infection rate of sheep in different regions and different breeds was not significantly different.There was a risk of zoonosis of Giardia duodenalis in Yunnan province.
