摘  要:目的探讨黏附素乙酰转移酶Esco2对肺癌细胞增殖和迁移的影响及可能机制。方法通过查找TCGA数据库中Esco2基因在肺癌中的差异表达与临床分期、生存率的关系。应用Esco2特异性siRNA干扰肺癌细胞系A549和H460,应用Transwell、MTX和集落形成实验检测Esco2对细胞增殖和侵袭能力的影响,应用蛋白印迹检测干扰Esco2后对CDK1、CDK4、c-myc及p53蛋白表达水平变化。结果Esco2基因在肺癌组织的表达水平明显高于癌旁组织,其表达与临床分期和生存率呈负相关(P<0.05),与P53呈负相关。特异性干扰Esco2后,细胞的增殖能力、集落形成能力和迁移能力均明显下调。蛋白印迹显示特异性干扰Esco2后,CDK1及c-myc蛋白水平明显下调,而p53蛋白水平上调(P<0.05)。结论Esco2促进肺癌细胞的增殖和迁移能力,与上调CDK1、c-myc及下调p53蛋白水平相关。Objective To investigate the effect and possible mechanism of Esco2 on the proliferation,migration of lung cancer cells.Methods The expression of Esco2 in lung cancer cells was investigated and its relationship with gene difference and survival rate and clinical stages was analyzed using TCGA database.Protein expression levels of Esco2 were regulated in non-small cell lung cancer cell lines A549 and H1299 by specific siRNA interference,and verified by immunoblotting.On this basis,the MTX was used to detect the effect of Esco2 expression on proliferation of lung cancer cells.The colony formation was used to detect the influence of Esco2 expression on the colony formation abilities of lung cancer cells.The Transwell assay was performed to explore the effect of Esco2 expression on the migration abilities of lung cancer cells.Meanwhile,the expressions of CDK1,c-myc and p53 protein expression were detected by western blot.Results Esco2 is highly expressed in cancer tissues,correlated with poor prognosis and clinical stages.Esco2 knock-down inhibits the proliferation and colony formation and migration of lung cancer cells,and down-regulates the levels of CDK1 and c-myc,but up-regulates p53 expression.Conclusion Esco2 plays a critical role in promoting the malignancy of lung cancer.
