补中益气汤含药血清对A549/DDP细胞P53、Bax、Bcl-2表达的影响

摘　　要：目的探讨Snail与凋亡因子P53、Bax、Bcl-2的相关性及补中益气汤含药血清对A549/DDP细胞P53、Bax、Bcl-2表达的影响。方法20只SD大鼠,随机分成两组,分别给予将补中益气汤及生理盐水灌胃处理,采用血清药理学研究方法制备含药血清。细胞分组:TGF-β_(1)(-)组、TGF-β_(1)(+)组(5ng/mL TGF-β_(1),作用48h)、TCM组(10%最佳含药血清组)、Snail-si组(Snail干扰组)。Western blot与Real time PCR方法分别检测各组细胞P53、Bax、Bcl-2基因及蛋白表达水平。结果与TGF-β_(1)(-)组比较,TGF-β_(1)(+)组P53、Bax蛋白及mRNA下调,Bcl-2 mRNA及蛋白表达上调。TCM组与TGF-β_(1)(+)组比较,P53、Bax蛋白及mRNA表达量增加,Bcl-2蛋白及mRNA表达量减少(P<0.05)。结论A549/DDP细胞Snail与凋亡因子P53、Bax、Bcl-2具有一定的相关性,表明补中益气汤含药血清可通过调节A549/DDP细胞Snail表达水平,干预凋亡相关因子的表达。Objective To explore the correlation of snail and P53,Bax,Bcl-2,and observe the effects of Buzhong Yiqi decoction on the expressions of P53,Bax and Bcl-2 on cisplatin-resistant cell line A549/DDP.Methods Twenty SD rats were randomly divided into two groups,which were treated with Buzhong Yiqi decoction and normal saline by gavage respectively,and the drug-containing serum was prepared by serum pharmacology.The experiment contain four groups:TGF-β_(1)(-)group;TGF-β_(1)(+)group(5ng/mL TGF-β_(1),48h);TCM group(10%Buzhong Yiqi Decoction rat serum)、Snail-si group(Snail siRNA).Western blot and Real-time PCR methods were used to detect the expressions of P53,Bax,Bcl-2 protein and mRNA.Results Compare with TGF-β_(1)(-)groups,mRNA and protein expressions of P53 and Bax were significantly lower in TGF-β_(1)(+)groups,mRNA and protein expressions of Bcl-2 was higer in TGF-β_(1)(+)groups(P<0.05).Compare with TGF-β_(1)(+)groups,mRNA and protein expressions of P53 and Bax were significantly higer in TCM groups,mRNA and protein expressions of Bcl-2 was significantly lower in TCM groups(P<0.05).Conclusion The mRNA and protein expressions of P53、Bax、Bcl-2 were related to snail,serum containing Buzhong Yiqi decoction can interfere with the expression of apoptosis-related factors by regulating the expression level of Snail on A549DDP cells.
