摘 要:目的探讨冬凌草甲素对人骨肉瘤MG63细胞增殖与侵袭的影响及可能机制。方法将人骨肉瘤MG63细胞分为对照组、冬凌草甲素低、中、高剂量组。低、中、高剂量组MG63细胞分别用浓度10、20、40μmol/L的冬凌草甲素溶液处理,对照组MG63细胞采用等量二甲基亚砜(DMSO)处理。MTT实验检测各组细胞增殖能力;流式细胞仪检测各组细胞凋亡率;Transwell实验检测各组细胞的侵袭能力;Western blot实验和实时荧光定量PCR方法检测各组细胞mTOR、P70S6K蛋白及mRNA的表达。结果与对照组相比,低、中、高剂量的冬凌草甲素溶液处理MG63细胞后,细胞的增殖与侵袭能力明显降低,凋亡率明显升高,细胞中mTOR与P70S6K的蛋白及mRNA表达水平均明显降低(P<0.05)。结论冬凌草甲素能够通过抑制mTOR/P70S6K信号通路促进MG63细胞凋亡,并抑制增殖与侵袭。Objective To investigate the effect of oridonin on proliferation and invasion of human osteosarcoma MG63 cells and its possible mechanism. Methods Human osteosarcoma MG63 cells were divided into control group, low, medium and high dose oridonin groups. MG63 cells in low, medium and high dose groups were treated with 10, 20 and 40 μmol/L of oridonin solution, respectively, while MG63 cells in control group were treated with the same amount of dimethyl sulfoxide(DMSO). The proliferation of MG63 cells of each group was detected by MTT assay. The apoptosis rate of MG63 cells of each group was detected by flow cytometry. The invasion of MG63 cells in each group was detected by Transwell assay. The expressions of mTOR and P70S6K protein and mRNA were detected by Western blot and qRT-PCR, respectively. Results Compared with the control group, the proliferation and invasion ability of MG63 cells in low, medium and high dose oridonin groups were significantly decreased, the apoptosis rate was significantly increased, and the protein and mRNA expression levels of mTOR and P70S6K in the cells were significantly decreased(P<0.05). Conclusion Oridonin could promote apoptosis, inhibit proliferation and invasion of MG63 cells by inhibiting mTOR/P70S6K signaling pathway.
