摘  要:目的 探讨鱼腥草素对非小细胞肺癌细胞(NSCLCs)增殖的影响和机制。方法 将NSCLCs分为对照组(Control)、低、中及高剂量鱼腥草素组。CCK8检测细胞存活率;克隆形成实验检测细胞克隆能力;流式细胞术检测细胞凋亡率和细胞周期;Western blot检测NCI-H1734细胞中EGFR、AKT和ERK以及磷酸化表达水平。结果 鱼腥草素抑制A549、NCI-H1734和NCI-H1299细胞存活率、减少细胞克隆形成数量、增加细胞凋亡率、减少G2/M期细胞数量、抑制NCI-H1734细胞中pEGFR、pAKT和pERK蛋白表达水平,且呈剂量依赖性。结论 鱼腥草素通过抑制EGFR信号通路磷酸化抑制NSCLCs增殖,促进凋亡和诱导周期阻滞。Objective To investigate the effect and mechanism of houttuynin on the proliferation of non-small cell lung cancer cells(NSCLCs). Methods The NSCLCs were divided into control group, low, medium and high dose houttuynin group. Cell survival rate was detected by CCK8. The clonal ability of cells was detected by clonal formation assay. The apoptosis rate and cycle of cells were detected by flow cytometry. The expressions of EGFR, AKT, ERK and phosphorylation were detected by Western blot. Results Houttuynin inhibited the survival rate of A549, NCI-H1734 and NCI-H1299 cells, decreased the number of cell clones, increased the apoptosis rate, decreased the number of G_2/M phase cells, and inhibited the expressions of pEGFR, pAKT and pERK proteins in NCI-H1734 cells in a dose-dependent manner. Conclusion Houttuynia inhibits NSCLCs proliferation, promotes apoptosis and induces cycle arrest by inhibiting EGFR signaling pathway phosphorylation.
