摘 要:目的 探讨柴胡皂苷D通过糖酵解途径对乳腺癌细胞增殖的影响。方法 将浓度为0μmol/L、10μmol/L、20μmol/L和50μmol/L的柴胡皂苷D加入到乳腺癌细胞中,通过四甲基偶氮唑盐法(tetramethylazolium salt, MTT)和免疫蛋白印迹法(Western blot)分别检测MCF-7细胞活力和细胞中STAT3、JAK2、p-STAT3和p-JAK2蛋白表达水平;试剂盒检测分析细胞凋亡变化和乳酸生成和葡萄糖消耗情况;qPCR检测MCF-7细胞糖酵解相关酶mRNA水平。结果 不同时间点,10μmol/L、20μmol/L和50μmol/L柴胡皂苷D均能使MCF-7细胞活力显著降低(P<0.05);细胞凋亡率均升高(P<0.05);降低MCF-7细胞葡萄糖消耗量,乳酸含量以及相关酶mRNA表达(P<0.05);降低细胞中p-JAK2和p-STAT3相对表达量(P<0.05)。结论 柴胡皂苷D可能通过抑制JAK2/STAT3通路抑制乳腺癌细胞增殖,促进细胞凋亡,抑制细胞糖酵解过程。Objective To investigate the effect of saikosaponins D on the proliferation of breast cancer cells through glycolytic pathway. Methods Saikosaponin D at concentrations of 0 μmol/L, 10 μmol/L, 20 μmol/L and 50 μmol/L were added to breast cancer cells, the viability of MCF-7 cells and the expressions of STAT3, JAK2, p-STAT3 and p-JAK2 in MCF-7 cells were detected by tetramethylazolium salt(MTT)and Western blot. The changes of apoptosis, lactate production and glucose consumption were detected by kits. The mRNA levels of glycolysis-related enzymes in MCF-7 cells were detected by qPCR. Results 10 μmol/L, 20 μmol/L and 50 μmol/L saikosaponin D significantly reduced MCF-7 cell viability(P<0.05);increased apoptosis rate of MCF-7 cells(P<0.05);reduced glucose consumption, lactic acid content and mRNA expressions of related enzymes in MCF-7 cells(P<0.05);reduced the relative expressions of p-JAK2 and p-STAT3 in cells(P<0.05). Conclusion Saikosaponin D can inhibit the proliferation of breast cancer cells, promote cell apoptosis and inhibit the process of cell glycolysis by inhibiting JAK2/STAT3 pathway.
