青藤碱调控COX-2/VEGF通路抑制肝癌HepG2细胞增殖和侵袭

摘　　要：目的探讨青藤碱能否通过调控COX-2/VEGF通路抑制肝癌HepG2细胞增殖和侵袭。方法体外常规培养HepG2细胞,随机将HepG2细胞分为0、1.0、2.0 mmol/L青藤碱处理组。MTT实验、流式细胞术与Transwell实验分别检测青藤碱对人肝癌HepG2细胞增殖、凋亡与侵袭能力的影响;Western blot实验和实时荧光定量PCR实验检测青藤碱对人肝癌HepG2细胞COX-2与VEGF蛋白与mRNA表达的影响。结果青藤碱能够降低人肝癌HepG2细胞的增殖活力,促进凋亡,并抑制人肝癌HepG2细胞的体外侵袭能力;降低人肝癌HepG2细胞COX-2与VEGF mRNA和蛋白表达(P<0.05)。结论青藤碱通过抑制COX-2/VEGF通路抑制肝癌HepG2细胞增殖和侵袭,促进凋亡。Objective To investigate whether sinomenine can inhibit the proliferation and invasion of hepatocellular carcinoma HepG2 cells by regulating COX-2/VEGF pathway.Methods HepG2 cells were routinely cultured in vitro and randomly divided into 0,1.0 and 2.0 mmol/L sinomenine treatment group.MTT assay,flow cytometry assay and transwell assay were used to detect the effects of sinomenine on the proliferation,apoptosis and invasion of human hepatocellular carcinoma HepG2 cells.The effects of sinomenine on the expressions of COX-2 and VEGF protein and mRNA in human hepatocellular carcinoma HepG2 cells were detected by Western blot and real-time fluorescence quantitative PCR.Results Sinomenine can decrease the proliferation activity,promote apoptosis and inhibit the invasion ability of HepG2 cells in vitro.The mRNA and protein expressions of COX-2 and VEGF in human hepatoma HepG2 cells were decreased(P<0.05).Conclusions Sinomenine inhibits proliferation and invasion of hepatoma HepG2 cells and promotes apoptosis by inhibiting COX-2/VEGF pathway.
