摘 要:目的探究大黄酚对EGF诱导的甲状腺乳头状癌TPC-1细胞增殖、迁移、侵袭和EMT的抑制作用,并探讨其可能机制。方法TPC-1细胞分为对照组、EGF组、大黄酚低剂量组、大黄酚中剂量组和大黄酚高剂量组。采用CCK-8法测定细胞增殖活性,Transwell实验检测细胞迁移和侵袭能力,Western blot测定细胞EMT相关蛋白N-cadherin和Vimentin以及Claudin 1蛋白表达及EGFR磷酸化水平。结果EGF处理增加TPC-1细胞增殖、迁移和侵袭能力,上调细胞中N-cadherin、Vimentin、p-EGFR和Claudin 1蛋白表达水平。大黄酚处理剂量依赖的抑制TPC-1细胞增殖、迁移和侵袭能力,降低细胞中N-cadherin、Vimentin、p-EGFR和Claudin 1蛋白表达水平。结论大黄酚抑制了EGF诱导的PTC细胞增殖、迁移、侵袭和EMT,其机制可能与抑制EGFR激活并下调Claudin 1表达有关。Objective To explore the inhibitory effect of chrysophanol on EGF-induced proliferation,migration,invasion and EMT of thyroid papillary carcinoma TPC-1 cells,and to explore its possible mechanism.Methods TPC-1 cells were divided into control group,EGF group,low-dose,medium-dose and high-dose chrysophanol group.Cell proliferation activity was detected by CCK-8 assay,cell migration and invasion ability was detected by Transwell assay,and the expressions of EMT-related proteins N-cadherin,Vimentin,Claudin 1 and EGFR phosphorylation levels were determined by Western blot.Results EGF treatment increased the proliferation,migration and invasion ability of TPC-1 cells,and up-regulated the expression levels of N-cadherin,Vimentin,p-EGFR and Claudin 1 proteins in TPC-1 cells.Chrysophanol treatment inhibited the proliferation,migration and invasion of TPC-1 cells in a dose-dependent manner,and decreased the expression levels of N-cadherin,Vimentin,p-EGFR and Claudin 1 proteins in the cells.Conclusion Chrysophanol inhibited EGF-induced proliferation,migration,invasion and EMT of PTC cells,and the mechanism may be related to inhibiting EGFR activation and down-regulating Claudin 1 expression.
