摘 要:采用水热法制备合成UiO-66金属有机框架,通过回流搅拌法将Ru(bpy)_(3)^(2+)嵌入UiO-66中形成复合材料UiO-Ru.应用粉末X射线衍射(PXRD)、扫描电子显微镜(SEM)、粒径与zeta电位分析确定其结构,采用电感耦合等离子体发射光谱仪(ICP-OES)、紫外吸收光谱和荧光发射光谱等对UiO-Ru进行表征.在钌配合物修饰后,UiO-Ru的结构保持不变.Ru(bpy)_(3)^(2+)的加入使UiO-Ru具有一定的光活性且光稳定性显著增强.在650nm光照下,UiO-Ru能够有效产生单线态氧,其细胞暗毒性低,在宫颈癌细胞(HeLa细胞)中表现出明显的光毒性和良好的光动力治疗效果,为金属有机框架材料应用于光动力治疗提供了新思路.UiO-66 metal-organic frameworks are prepared by hydrothermal method,and Ru(bpy)_(3)^(2+)is embedded in UiO-66 by reflux stirring method to form the composite UiO-Ru.The structure of UiO-Ru is determined by powder X-ray diffraction(PXRD),scanning electron microscope(SEM),particle size and zeta potential analysis,and UiO-Ru is characterized by inductively coupled plasma optical emission spectrometry(ICP-OES),UV-absorption spectroscopy and fluorescence emission spectroscopy.The structure of UiO-Ru remains unchanged after the modification of the ruthenium complexes,and the incorporation of Ru(bpy)_(3)^(2+) confers certain photoactivity to UiO-Ru and significantly enhances its photostability.UiO-Ru with low dark cytotoxicity can effectively produce singlet oxygen in the light of 650 nm,and exhibits significant photocytotoxicity and good photodynamic therapy effect in cervical cancer cells(HeLa cells),which provides a new idea for metal-organic frameworks materials applied to photodynamic therapy.
