摘 要:将结扎兔心脏冠状动脉左前降支(left anterior descending branch,LADB)和左室支(left ventricular branch,LVB)建立心肌缺血再灌注损伤(ischemia-reperfusion injury,IRI)模型的两种实验方法进行比较,为从事相关领域的科学研究提供模型建立方法参考.将42只新西兰大白兔随机分组后,在呼吸机辅助下,离断左侧第3-4肋间肌暴露心脏,分别定位结扎LADB或LVB建立IRI模型.研究发现,与结扎LADB相比,结扎LVB兔心电图演变典型、血清肌钙蛋白cTnI含量明显升高、心肌组织p-P38 MAPK蛋白表达量明显升高、组织学变化明显,心肌梗死面积明显增大.本研究认为,结扎LVB建立兔心肌缺血再灌注损伤模型方法明显优于结扎LADB.The left anterior descending branch(LADB)or left ventricular branch(LVB)of cardiac coronary artery was ligated in rabbits.Two experimental methods of establishing myocardial ischemia-reperfusion injury(IRI)model were compared to provide the reference for scientific research in related fields.42 New Zealand white rabbits were randomly divided into four groups,the left 3-4 intercostal muscle was severed to expose the heart with the aid of ventilator,and the IRI model has been established by the ligation of LADB or LVB.Comparing with LADB ligation group,the electrocardiogram changed obviously,serum troponin c Tn I levels increased significantly,the expression of myocardial p-P38 MAPK protein increased significantly,the histological changed obviously and myocardial infarction area significantly increased of the rabbits suffering LVB ligation group.The LVB ligation has been superior to LADB ligation to establish rabbit myocardial ischemia-reperfusion injury model in this study.
