摘  要:目的探讨葛根素(Pue)调节丝裂原活化蛋白激酶(MAPK)/核转录因子-κB(NF-κB)信号通路对大鼠正畸牙移动过程中牙周组织的影响。方法取SD大鼠随机分为对照组(C组)、模型组(M组)、葛根素低剂量(Pue-L组)、葛根素高剂量(Pue-H组)、葛根素(200 mg/kg)+茴香霉素(ANS)组(Pue+ANS组),每组12只,除C组外,其余各组大鼠建立正畸牙模型,然后以葛根素和茴香霉素分组干预各组大鼠后检测其牙齿移动距离、牙槽骨骨密度、骨微结构、血清白细胞介素(IL)-1β、IL-6、肿瘤坏死因子-α(TNF-α)、骨保护素(OPG)、核因子κB受体活化因子配体(RANKL)及单核细胞趋化性蛋白-1(MCP-1)水平,检测牙周组织中IL-1β、TNF-α、MCP-1、IL-6水平及MAPK/NF-κB通路相关蛋白表达。结果与C组相比,M组大鼠牙槽骨骨微结构受损,牙齿移动距离、血清IL-1β、TNF-α、IL-6、MCP-1及RANKL水平、p-p38 MAPK/p38 MAPK、p-NF-κB p65/NF-κB p65增高(P<0.05);与M组相比,Pue-L、Pue-H组大鼠牙齿移动距离、牙槽骨骨密度、骨体积分数(BV/TV)、骨小梁数目(Tb.N)、骨小梁厚度(Tb.Th)、血清OPG水平均升高(P<0.05),骨小梁间隙(Tb.Sp)、血清IL-1β、TNF-α、IL-6、MCP-1及RANKL水平、p-p38 MAPK/p38 MAPK、p-NF-κB p65/NF-κB p65均降低,Pue-H组大鼠上述各指标变化比Pue-L组更强;与Pue-H组相比,Pue+ANS组大鼠牙齿移动距离、牙槽骨骨密度、骨体积分数(BV/TV)、骨小梁数目(Tb.N)、骨小梁厚度(Tb.Th)、血清OPG水平均降低(P<0.05),骨小梁间隙(Tb.Sp)、血清IL-1β、TNF-α、IL-6、MCP-1及RANKL水平、p-p38 MAPK/p38 MAPK、p-NF-κB p65/NF-κB p65均升高(P<0.05)。结论Pue可抑制MAPK/NF-κB信号激活,进而减少炎性因子释放,抑制正畸牙模型大鼠牙周组织炎症,减轻其牙槽骨骨量丢失及微结构损伤,并加速正畸牙移动。Objective To investigate the effects of puerarin(Pue)on periodontal tissue during orthodontic tooth movement in rats by regulating the mitogen-activated protein kinase(MAPK)/nuclear transcription factor-κB(NF-κB)signaling pathway.Methods SD rats were randomly divided into a control group(C group),a model group(M group),a low-dose puerarin group(Pue-L group),a high-dose puerarin group(Pue-H group),and a puerarin(200mg/kg)+anisomycin group(ANS)(Pue+ANS group),12 rats in each group.Except for the control group,orthodontic tooth models were established in all groups,and then intervened with puerarin and/or anisomycin.After intervention,the distance of tooth movement,bone mineral density(BMD),bone microstructure,levels of serum interleukin-1β(IL-1β),IL-6,tumor necrosis factor-α(TNF-α),osteoprotegerin(OPG),receptor activator of nuclear factor-κB ligand(RANKL)and monocyte chemotactic protein-1(MCP-1)were detected.The levels of IL-1β,TNF-α,MCP-1 and IL-6 and the expression of MAPK/NF-κB pathway related proteins in periodontal tissue were also detected.Results Compared with the group C,rats in the group M had damage to the alveolar bone microstructure,tooth movement distance,serum IL-1β,TNF-α,IL-6,MCP-1 and RANKL levels,P-p38 MAPK/p38 MAPK,p-NF-κB p65/NF-κB p65 were increased(P<0.05).Compared with the M group,the tooth movement distance,alveolar bone density,bone volume fraction(BV/TV),trabecular number(Tb.N),trabecular thickness(Tb.Th),and serum OPG levels were increased(P<0.05),and trabecular space(Tb.Sp),serum IL-1β,TNF-α,IL-6,MCP-1 and RANKL levels,P-p38 MAPK/p38 MAPK,and p-NF-κB p65/NF-κB p65 were all reduced in the Pue-H group and the Pue-L group,and the changes in the above indicators of rats in the Pue-H group were stronger than those in the Pue-L group.Compared with the Pue-H group,the tooth movement distance,alveolar bone BMD,such as bone volume/tissue volume(BV/TV),Tb.N,Tb.Th,and serum OPG level were significantly decreased(P<0.05)while Tb.SP,serum levels of IL-1β,TNF-α,IL-6,MCP-1 and RANKL
