摘  要:核孔复合物由核孔蛋白组成,位于细胞核膜上,为细胞质与细胞核之间的关键物质输送通道,介导小分子物质、大分子蛋白、mRNA等物质的输送。现有研究表明,部分核孔蛋白在肿瘤中表达异常,通过影响PI3K/AKT/mTOR、TGF-β/SMAD信号通路,YAP、β-catenin核转位,调节肿瘤的增殖、凋亡、细胞周期、迁移、侵袭等,影响肿瘤的进展。核孔蛋白Nup98的融合表达与其他基因的突变联合导致癌症的发生。本文主要阐述了组成核孔复合物的关键核孔蛋白分布,核孔蛋白调控细胞核与细胞质之间的物质转运,以及核孔蛋白与肿瘤发生发展之间的关系。Nuclear pore complex,located on the nuclear membrane,is a key material transport channel between the cytoplasm and the nucleus.It mediates the entry and exit of small molecular substances,macromolecular proteins,mRNA,and other substances.According to recent research,several nucleoporins were abnormally expressed in tumors and regulate tumor progression by affecting the PI3K/AKT/mTOR and TGF-β/SMAD signaling pathways,YAP andβ-catenin nuclear translocation.Nucleoporins regulate tumor proliferation,apoptosis,cell cycle,migration,and invasion through these pathways.Nucleoporins fusion expression of the Nup98 combined with mutations of other genes may lead to cancer occurrence.This article primarily reviewed the distribution of key nucleoporins that constitute the nuclear pore complex.Nucleoporins regulated the transport of substances between the nucleus and the cytoplasm.Furthermore,the relationship between nucleoporins and tumorigenesis and tumor progression was also discussed.
