摘  要:目的:探讨不同临床分期及位置的食管鳞状细胞癌(鳞癌)菌群组成及差异性。方法:选择食管鳞癌30例,其中临床分期Ⅰ期8例、Ⅱ期6例、Ⅲ期4例、Ⅳ期6例、未分期6例,肿瘤位置食管上段4例、中段19例、下段5例、无法判断位置2例。采用16S rDNA基因测序技术检测食管鳞癌组织的菌群组成。通过主成分分析和线性判别分析法评估不同临床分期和位置食管鳞癌菌群结构和组成的差异。结果:临床Ⅰ期和Ⅱ期食管鳞癌的菌群结构具有高度相似性,与Ⅲ期和Ⅳ期差异明显;不同位置的食管鳞癌菌群结构差异较大。假单胞菌和普雷沃菌的相对丰度在临床Ⅰ~Ⅳ期食管鳞癌组织呈上升趋势,食管上段到下段呈下降趋势。链球菌在临床Ⅰ期和Ⅱ期的相对丰度高于Ⅲ期和Ⅳ期,食管中段高于上段和下段。结论:假单胞菌、普雷沃菌和链球菌可能与食管鳞癌的发生发展有关。Aim:To investigate the composition and differences of the microbiota in esophageal squamous cell carcinoma(ESCC)at different clinical stages and tumor locations.Methods:A total of 30 ESCC cases were selected,including 8 cases at clinical stageⅠ,6 at stageⅡ,4 at stageⅢ,6 at stageⅣ,and 6 unclassified.The tumor locations included 4 in the upper esophagus,19 in the middle esophagus,5 in the lower esophagus,and 2 unclassified.The composition of the microbiota in ESCC tissue was detected using 16S rDNA gene sequencing.Principal component analysis and linear discriminant analysis were used to assess differences in microbiota structure and composition of ESCC at different clinical stages and tumor locations.Results:The microbiota structure of ESCC at stagesⅠandⅡwas similar;however,it differed significantly from that of stagesⅢandⅣ.The microbiota composition varied greatly across different tumor locations.The relative abundance of Pseudomonas and Prevotella showed an increasing trend from clinical stagesⅠtoⅣ,but a decreasing trend from the upper to the lower esophagus.The relative abundance of Streptococcus was higher in ESCC at clinical stagesⅠandⅡthan that at stagesⅢandⅣ,and higher in the middle esophagus than that in the upper or lower esophagus.Conclusion:Pseudomonas,Prevotella,and Streptococcus may be associated with the development and progression of ESCC.
