摘　　要：目的:探讨T细胞免疫球蛋白黏蛋白3(TIM3)、程序性死亡受体1(PD-1)与程序性死亡受体配体1(PD-L1)在子宫内膜样腺癌组织中的表达及意义。方法:选取2021年1月至2022年12月行手术切除的50例子宫内膜样腺癌患者的癌、癌旁子宫内膜非典型增生和正常子宫内膜组织。采用免疫组化检测TIM3、PD-1和PD-L1蛋白的表达,qRT-PCR法检测TIM3、PD-1和PD-L1 mRNA的表达,分析子宫内膜样腺癌组织中TIM3、PD-1和PD-L1 mRNA表达的相关性。比较不同临床特征的子宫内膜样腺癌患者癌组织中TIM3、PD-1和PD-L1表达的差异。结果:3种组织中TIM3、PD-1、PD-L1蛋白和mRNA表达的差异有统计学意义,子宫内膜非典型增生和子宫内膜样腺癌组织高于正常子宫内膜组织,子宫内膜样腺癌组织高于子宫内膜非典型增生组织(P<0.05);TIM3 mRNA与PD-1、PD-L1 mRNA的表达呈正相关(r P=0.471和0.512,P<0.001);有淋巴结转移,TNM分期Ⅲ期和中、低分化程度的子宫内膜样腺癌患者癌组织中TIM3、PD-1蛋白表达的阳性率较高(P<0.05)。结论:TIM3、PD-1、PD-L1可能与子宫内膜样腺癌的发生发展有关。Aim:To explore the expressions of T cell immunoglobulin and mucin domain-containing protein 3(TIM3),programmed death-1(PD-1),and programmed death-ligand 1(PD-L1)in endometrioid adenocarcinoma tissues and their clinical significance.Methods:Endometrioid adenocarcinoma,corresponding atypical hyperplasia and normal endometrial tissues were selected from 50 patients with endometrioid adenocarcinoma who underwent surgical treatment from January 2021 to December 2022.Immunohistochemistry was used to detect the expressions of TIM3,PD-1,and PD-L1 protein,and qRT-PCR was used to detect the expressions of TIM3,PD-1,and PD-L1 mRNA.The correlation between the expressions of TIM3,PD-1,and PD-L1 mRNA in endometrioid adenocarcinoma tissue was analyzed.The expressions of TIM3,PD-1,and PD-L1 in endometrioid adenocarcinoma tissue of the patients with different clinical characteristics were compared.Results:There were statistically significant differences in the expressions of TIM3,PD-1,and PD-L1 protein and mRNA among the 3 types of tissue,which were higher in endometrial atypical hyperplasia and endometrioid adenocarcinoma tissue than those in normal endometrial tissue,and in endometrioid adenocarcinoma tissue than those in endometrial atypical hyperplasia tissue(P<0.05).The expression of TIM3 mRNA was positively correlated with PD-1 and PD-L1 mRNA(r P=0.471 and 0.512,P<0.001).The positive rates of TIM3 and PD-1 protein expressions in endometrioid adenocarcinoma tissue of patients with lymph node metastasis,TNM stageⅢ,and moderate to low differentiation were higher(P<0.05).Conclusion:TIM3,PD-1,and PD-L1 may be related to the occurrence and development of endometrioid adenocarcinoma.
