培养基种类及IBA浓度对隔山消不定根生长及生物活性物质积累的影响

摘  要:为了筛选出隔山消不定根培养的适宜培养基种类和吲哚丁酸(IBA)浓度,该研究探究了五种基本培养基种类(MS、SH、WPM、White、B_(5))和不同IBA浓度(0.5、1、1.5、2、2.5mg·L^(-1))对隔山消不定根生长和C_(21)甾苷、多糖和多肽积累的影响。结果表明:在五种培养基种类中,隔山消不定根生物量、三种生物活性物质(C21甾苷、多糖和多肽)含量及其生产量在MS培养基中均最高。在IBA浓度筛选试验中发现,添加IBA浓度为1.5mg·L^(-1)时隔山消不定根的生物量鲜重(22.2g)和生物量干重(1.7g)均达到最高,多糖及多肽含量也达到均最高,分别为168.0mg·g^(-1)DW和80.4mg·L^(-1)DW,此浓度下多糖和多肽生产量分别达到2889.2mg·L^(-1)和1383.1mg·L^(-1);然而C_(21)甾苷积累在IBA2mg·L^(-1)时最佳,其含量为145.6mg·L^(-1),生产量达到2460.8mg·L^(-1)。综合考虑下,培养隔山消不定根的培养基选为MS并加入1.5mg·L^(-1)IBA较适宜。To select a suitable medium type and indole butyric acid(IBA)concentrations for the adventitious root(AR)culture of Cynanchum wilfordi,this study investigated the effects of five basic medium types(MS,SH,WPM,White,B_(5))and different IBA concentrations(0.5,1,1.5,2,2.5 mg·L^(-1))on AR growth and accumulation of Cai steroidal glycosides,polysaccharides,and polypeptides.The results showed that among the five medium types,the AR biomass and the contents of three bioactive compounds and their productivities were highest in MS medium.In the IBA concentration selection experiment,AR fresh weight(22.2 g)or dry weight(DW)(1.7 g)reached the highest with the addition of IBA at a concentration of 1.5 mg·L^(-1),The polysac-charide and polypeptide contents also reached the highest at this IBA concentration,which were 168.0 mg·g^(-1) DW and 80.4 mg·g^(-1)DW,respectively At this concentration,the production of polysaccharides and polypep-tides reached 2889.2 mg·L^(-1) and 1383.1 mg·L^(-1).However,the accumulation of C21 steroidal glycosides was optimal at 2 mg L^(-1) IBA,with a content of 145.6 mg·L^(-1) DW and production of 2460.8 mg·L^(-1).In conclusion,MS medium supplemented with 1.5 mg·L^(-1) IBA can be selected for C.wilfordi AR culture by comprehensive evaluation.
