摘 要:本研究介绍了一种基于离子排斥色谱原理同时测定乳酸和7种短链脂肪酸的检测方法,并以肠道内容物和粪便为基质,对该方法进行方法学评价。结果表明:本试验方法测定的8种有机酸含量线性范围为1~500μg/mL时,其线性相关系数均>0.999,方法检出限为0.02~0.15 g/kg;在添加不同含量(1、4和10 g/kg)的8种有机酸标准品条件下,方法回收率为80.28%~116.99%,相对标准偏差均<10%。本方法前处理简单,可同时检测与动物肠道相关的多种关键有机酸含量,为短链脂肪酸和乳酸的检测提供了一种新的方法参考。In this study,a detection method for simultaneous determination of lactic acid and seven short-chain fatty acids based on the principle of ion exclusion chromatography was introduced,and the method was evaluated using intestinal contents and feces as substrates.The results showed that when the linear range of the contents of eight organic acids was from 1 to 500μg/mL,the linear correlation coefficients were all greater than 0.999,and the detection limit was from 0.02 to 0.15 g/kg.The recovery rates ranged from 80.28% to 116.99%,and the relative standard deviations were all smaller than 10%,when eight organic acid standard substances with different contents(1,4 and 10 g/kg)were added.This preprocessing of this method is simple and can simultaneously detect the contents of many key organic acids related to animal gut,providing a new method for the detection of short-chain fatty acids and lactic acid.
