摘　　要：目的:研究骨肉瘤组织锌-α2-糖蛋白1(AZGP1)表达变化,分析其与患者预后的关系。方法:选取骨肉瘤患者40例,以手术中切除的骨肉瘤组织及癌旁正常组织(距离肿瘤边缘至少5 cm)为标本,实时荧光定量PCR(RT-qPCR)检测AZGP1 mRNA表达,免疫组织化学染色检测AZGP1蛋白表达,比较骨肉瘤组织与癌旁正常组织AZGP1 mRNA和蛋白表达,分析AZGP1蛋白表达与不同临床病理特征患者的关系以及骨肉瘤患者预后的影响因素。结果:骨肉瘤组织AZGP1 mRNA相对表达量及蛋白阳性率高于癌旁正常组织(均P<0.05)。AZGP1蛋白高表达患者Enneking分期Ⅲ期和软组织浸润占比高于低表达患者(均P<0.05)。Logistic回归分析结果显示,Enneking分期Ⅲ期和AZGP1高表达是骨肉瘤患者预后的独立危险因素(均P<0.05)。结论:AZGP1在骨肉瘤组织中表达升高,是骨肉瘤患者预后的危险因素。Objective:To study the expression of zinc-α2-glycoprotein 1(AZGP1)in osteosarcoma and its relationship with prognosis.Methods:Forty patients with osteosarcoma were selected,and the osteosarcoma tissues and adjacent normal tissues(at least 5 cm away from the tumor margin)removed during surgery were used as samples.RT-qPCR was used to detect AZGP1 mRNA expression,and immunohistochemical staining was used to detect AZGP1 protein expression.The expression of AZGP1 mRNA and protein in osteosarcoma tissues and adjacent normal tissues were compared.The relationship between AZGP1 protein expression and patients with different clinicopathological characteristics and the influencing factors of prognosis in osteosarcoma patients were analyzed.Results:The relative expression level of AZGP1 mRNA and positive rate of AZGP1 protein in osteosarcoma tissues were higher than those in adjacent normal tissues(all P<0.05).The proportion of Enneking stageⅢand soft tissue infiltration in patients with high expression of AZGP1 protein were higher than those in patients with low expression(all P<0.05).Logistic regression analysis showed that Enneking stageⅢand high expression of AZGP1 were independent risk factors for the prognosis of osteosarcoma patients(all P<0.05).Conclusion:The expression of AZGP1 is increased in osteosarcoma tissue,which is a risk factor for the prognosis of osteosarcoma patients.
