摘 要:目前,对凤香型大曲中红心曲、槐瓤曲、青茬曲的研究鲜有报道.本研究以这三种大曲感官特征最为显著的中心区域为试验样本(编号为HXH、HRA、QCU),通过PacBio细菌全长测序、UHPLC-Q Exactive HF-X非靶向代谢组学分析及理化特性检测,比较其差异性和相关性.研究结果表明HXH的水分、酸度、淀粉含量最高,HRA的糖化力和发酵力最高,QCU的酯化力最高;共检出95个细菌属,其中HXH的优势菌为克罗彭斯特菌属(Kroppenstedtia,25.79%)、芽孢杆菌属(Bacillus,16.90%)和明串珠菌属(Leuconostoc,10.55%),HRA的优势菌为糖多孢菌属(Saccharopolyspora,43.93%)、克罗彭斯特菌属(Kroppenstedtia,20.80%)和芽孢杆菌属(Bacillus,13.44%),QCU的优势菌为芽孢杆菌属(Bacillus,53.90%)、葡萄球菌属(Staphylococcus,38.90%)和unclassified_c_Actinomycetia(5.50%);代谢组学共检测到17类3447种功能代谢物,筛选出249种显著差异代谢物(VIP>1,P<0.05),并富集到具体关键代谢通路10条.三种凤香型大曲中细菌群落、理化特性、代谢产物相互影响,进而影响大曲的品质.研究结果对通过调节优势细菌群落结构,优化大曲理化特性,提升大曲品质,完善大曲质量评价体系有重要的理论与实际意义.At present,there are few reports on the research of Fengxiangxing Daqu(Hongxinqu,Huairangqu and Qingchaqu).In this study,the three central regions with the most significant sensory characteristics of Daqu were used as test samples(numbered HXH,HRA,QCU),and their differences and correlations were compared by full-length sequencing of PacBio bacteria,UHPLC-Q Exactive HF-X non-targeted metabolomics analysis and physicochemical properties.The results showed that HXH had the highest moisture,acidity,and starch content,the highest saccharification and fermentation power in HRA,and the highest esterification power in QCU;A total of 95 bacterial genera were detected,among which the dominant bacteria of HXH were Kroppenstedtia(25.79%),Bacillus(16.90%)and Leuconostoc(10.55%),and the dominant bacteria of HRA were Saccharopolyspora(43.93%),Kroppenstedtia(20.80%)and Bacillus(13.44%),and the dominant bacteria in QCU were Bacillus(53.90%),Staphylococcus(38.90%)and unclassified_c_Actinomycetia(5.50%);The metabolome detected 3447 metabolites in 17 classes,screened 249 significantly different metabolites(VIP>1,P<0.05)and enriched to 10 specific key metabolic pathways.Bacterial communities,physicochemical properties,and metabolites in three kinds of Fengxiangxing Daqu interacted with each other,which in turn affected the quality of Daqu.The results of this study have important theoretical and practical significance for optimising the physicochemical properties of Daqu by adjusting dominant bacterial community structure,improving the quality of Daqu and improving the quality evaluation system of Daqu.
