基于网络药理学对妇科再造丸治疗气血虚证的机理研究

摘  要:目的运用网络药理学与分子对接探究妇科再造丸治疗气血虚证的作用机制。方法通过TCMSP等数据库筛选妇科再造丸活性成分,运用SwissTargetPrediction确定成分基因靶点。运用OMIM等数据库查找疾病靶点。取成分与疾病的交集靶点导入STRING数据库构建PPI网络,筛选核心靶点。将交集靶点导入DAVID数据库进行GO功能及KEGG通路分析。最后对主要活性成分和核心靶点进行分子对接。结果得到514个妇科再造丸活性成分及1489个成分靶点基因;筛选到气血虚证疾病靶点6337个,交集靶点843个。经PPI网络分析得到8个核心靶点。GO分析主要涉及生物过程1644条、细胞组分214条、分子功能397条;KEGG通路富集得到214条。分子对接结果表明主要活性成分与核心靶点的对接构象合理。结论妇科再造丸可通过癌症通路、钙信号通路、MAPK信号通路等通路治疗气血虚证。Objective Objective To study the mechanism of action of Fuke Zaizao Pill in treating qi and blood deficiency syndrome by network pharmacology and molecular docking.Methods The active components of Fuke Zaizao Pill were screened by using TCMSP and other databases,and the target genes of the components were determined by using Swiss Target Prediction.OMIM and other databases were used to search for disease targets.The intersection targets of components and diseases were introduced into STRING database to construct PPI network and screen core targets.The intersection targets were also imported into the DAVID database for GO function and KEGG pathway analysis.Finally,molecular docking was performed on the main active ingredients and core targets.Results A total of 514 active components and 1,489 target genes of Fuke Zaizao Pill were obtained.A total of 6,337 disease targets of Syndrome of deficiency of both vital energy and blood and as well as 843 intersection targets were screened.8 core targets were obtained by PPI network analysis.GO analysis mainly involved 1,644 biological processes,214 cellular components,and 397 molecular functions.KEGG pathway enrichment yielded 214 entries.Molecular docking showed that the docking conformation of the main active components with the core target was reasonable.Conclusion Fuke Zaizao Pill can be used to treat qi and blood deficiency syndrome through pathways such as cancer pathway,calcium signaling pathway,MAPK signaling pathway and etc.
