子宫内膜癌患者癌组织中miR-136-5p、SOX9的表达水平及临床意义

摘  要:目的 研究微小RNA(miR)-136-5p与性别决定区Y框蛋白9(SOX9)在子宫内膜癌(EC)患者癌组织中的表达水平、临床意义。方法 收集2018年3月至2020年3月108例在杭州市第一人民医院进行手术治疗的EC患者癌组织与癌旁组织。实时荧光定量聚合酶链反应(qRT-PCR)法测定组织中miR-136-5p、SOX9相对表达量;分析miR-136-5p、SOX9表达的相关性分析;分析EC癌组织miR-136-5p与SOX9表达与生存的关系;分析EC患者预后的影响因素。结果 EC癌组织miR-136-5p水平低于癌旁组织(P<0.001),SOX9水平高于癌旁组织(P<0.001)。癌组织中SOX9表达率高于癌旁组织(χ^(2)=53.042,P<0.001)。miR-136-5p与SOX9间存在结合位点,且两者在EC患者癌组织中呈负相关(r=-0.325,P<0.001)。浸润深度、FIGO分期、组织分化程度、淋巴结转移与癌组织中miR-136-5p与SOX9有关(P<0.001)且为EC的影响因素。癌组织miR-136-5p高表达利于EC患者生存(χ^(2)=6.142,P=0.013),而SOX9低表达利于EC患者生存(χ^(2)=5.656,P=0.017)。结论 EC患者癌组织中miR-136-5p低表达,SOX9高表达,二者呈负相关关系,且影响患者临床病理特征以及预后。Objective To investigate the expression levels and clinical significance of microRNA-136-5p and sex determining region Y-box protein 9(SOX9)in endometrial cancer(EC)patients.Methods From March 2018 to March 2020,108 EC patients who underwent surgical treatment in Hangzhou First People’s Hospital,Hangzhou were gathered,and their cancerous and adjacent tissues were collected.Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)method was applied to determine the relative expression levels of miR-136-5p and SOX9 in tissues.Analyze the correlation between miR-136-5p and SOX9 expression.Analyze the relationship between miR-136-5p and SOX9 expression with survival in EC cancer tissues.Analyze the influencing factors of prognosis in EC patients.Results The expression level of miR-136-5p in EC cancer tissue was lower than that in adjacent tissues(P<0.001),and the expression level of SOX9 was higher than that in adjacent tissues(P<0.001).The expression rate of SOX9 in cancer tissue was higher than that in adjacent tissues(c2=53.042,P<0.001).There was a targeted binding site between miR-136-5p and SOX9.According to Pearson’s analysis,there was a negative correlation between miR-136-5p and SOX9 in cancer tissues of EC patients(r=‒0.325,P<0.001).The expression of miR-136-5p and SOX9 in cancer tissues of EC patients affected infiltration depth,International Federation of Gynecology and Obstetrics(FIGO)staging,tissue differentiation degree,and lymph node metastasis(P<0.001)are influencing factors of EC.High expression of miR-136-5p in cancer tissue is beneficial for the survival rate of EC patients(c2=6.142,P=0.013),low expression of SOX9 is beneficial for the survival of EC patients(c2=5.656,P=0.017).Conclusion The expression of miR-136-5p is low and the expression of SOX9 is high in cancer tissues of EC patients,and the two are negatively correlated,and affect the clinical pathological characteristics and prognosis of patients.
