子宫内膜癌组织中miR-199b-5p、SLC7A5的表达及与病理参数和预后的关系

摘　　要：目的探究子宫内膜癌(EC)组织中微小RNA-199b-5p(miR-199b-5p)、大氨基酸转运蛋白1(SLC7A5)的表达及与病理参数和预后的关系。方法选取2019年3月至2024年3月延安市人民医院收治的82例EC患者为研究对象。qRT-PCR法、免疫组织化学法分别检测EC组织中miR-199b-5p、SLC7A5表达水平;分析EC组织中miR-199b-5p、SLC7A5表达与临床病理参数及EC患者预后的关系;EC患者预后的影响因素用多因素Cox回归分析。结果EC组织中miR-199b-5p表达水平显著低于癌旁组织,SLC7A5的阳性表达率显著高于癌旁组织(P<0.05)。不同FIGO分期、淋巴结转移、分化程度及肌层浸润深度的EC患者miR-199b-5p低表达、SLC7A5阳性表达差异显著(P<0.05)。miR-199b-5p高表达患者3年生存率高于miR-199b-5p低表达患者;SLC7A5阳性表达患者3年生存率低于SLC7A5阴性表达患者。miR-199b-5p与SLC7A5呈负相关(P<0.05)。miR-199b-5p、SLC7A5、FIGO分期、淋巴结转移、肌层浸润深度是EC患者预后的影响因素(P<0.05)。结论EC组织miR-199b-5p表达水平降低、SLC7A5阳性表达率升高,且与EC患者临床病理参数及预后密切相关,对改善预后有一定临床应用价值。Objective To investigate the expression of microRNA-199b-5p(miR-199b-5p)and large-type amino acid transporter 1(SLC7A5)in endometrial cancer(EC)tissue and their relationship with pathological parameters and prognosis.Methods Eighty-two EC patients admitted to Yan’an People’s Hospital from March 2019 to March 2024 were regarded as the study subjects.QRT-PCR and immunohistochemistry were applied to detect the expression levels of miR-199b-5p and SLC7A5 in EC tissues,respectively.The relationship between the expression levels of miR-199b-5p and SLC7A5 in EC tissues with clinical pathological parameters and the prognosis of EC patients was analyzed.The factors influencing the prognosis of EC patients were analyzed using multivariate Cox regression.Results The expression level of miR-199b-5p in EC tissues was greatly lower than that in adjacent tissues,and the positive expression rate of SLC7A5 was greatly higher than that in adjacent tissues(P<0.05).There were significant differences in miR-199b-5p low expression and SLC7A5 positive expression in EC patients with different IGO stages,lymph node metastasis,differentiation degree and depth of myographic infiltration(P<0.05).The 3-year survival rate of patients with high expression of miR-199b-5p in EC tissue was higher than that of patients with low expression of miR-199b-5p;the 3-year survival rate of patients with SLC7A5 positive expression in EC tissue was lower than that of patients with SLC7A5 negative expression.miR-199b-5p was negatively correlated with SLC7A5(P<0.05).miR-199b-5p,SLC7A5,FIGO staging,lymph node metastasis,and depth of muscle infiltration were prognostic factors for EC patients(P<0.05).Conclusion The expression level of miR-199b-5p in EC tissue decreases,and the positive expression rate of SLC7A5 increases,and they are closely related to the clinical pathological parameters and prognosis of EC patients,which have certain clinical application value in improving prognosis.
