摘　　要：目的探讨子宫内膜癌(EC)患者血清中微RNA-429(miR-429)、DNA损伤诱导转录本4(DDIT4)表达水平及临床意义。方法纳入2020年5月至2021年5月石家庄市中医院收治EC患者(124例)作为观察对象(EC组),根据患者3年生存情况,将其分为生存组(79例)与死亡组(45例),另选择同期到石家庄市中医院体检的门诊健康者(120例)作为对照组。利用实时荧光定量PCR(qRT-PCR)检测血清miR-429表达水平,酶联免疫吸附试验(ELISA)检测血清DDIT4表达水平;采用Pearson法进行相关性分析;利用Kaplan-Meier生存曲线分析miR-429和DDIT4与患者预后相关性;利用Cox回归模型分析EC患者预后影响因素。结果EC组血清中miR-429表达水平低于对照组(P<0.05),DDIT4表达水平高于对照组(P<0.05)。Pearson相关性显示miR-429、DDIT4表达呈负相关(P<0.05)。EC患者血清miR-429、DDIT4表达水平与淋巴结转移阳性、浸润程度≥1/2cm有关(P<0.05)。血清miR-429低表达患者生存率(36.67%)显著低于miR-429高表达患者(89.06%)(P<0.05),DDIT4高表达患者生存率(35.48%)显著低于DDIT4低表达患者(91.94%)(P<0.05)。肌壁浸润程度≥1/2 cm、淋巴结转移阳性、miR-429低表达、DDIT4高表达是EC患者预后死亡的影响因素(P<0.05)。结论子宫内膜癌患者血清miR-429低表达,DDIT4高表达,与EC患者预后密切相关。Objective To investigate the expression levels and clinical significance of microRNA-429(miR-429)and DNA damage-inducible transcript 4(DDIT4)in the serum of patients with endometrial carcinoma(EC).Methods A total of 124 EC patients admitted to Shijiazhuang Hospital of Traditional Chinese Medicine from May 2020 to May 2021 were selected as the observation subjects(EC group).Based on the 3-year survival status of the patients,they were grouped into a survival group(79 cases)and a death group(45 cases).During the same period,120 cases of outpatient patients who underwent physical examination at Shijiazhuang Hospital of Traditional Chinese Medicine were included as the control group.Real-time fluorescence quantitative PCR(qRT-PCR)was applied to detect the expression level of miR-429.ELISA method was applied to detect the expression level of serum DDIT4.Pearson method was applied for correlation analysis.Kaplan-Meier survival curve was adopted to evaluate the correlation between miR-429 and DDIT4 with prognosis.Cox regression model was applied to analyze the prognostic factors affecting EC patients.Results The expression level of serum miR-429 in the EC group was lower than that in the control group(P<0.05),while the DDIT4 expression level was higher than that of the control group(P<0.05).Pearson correlation showed a negative correlation between miR-429 and DDIT4 expression(P<0.05).The expression levels of miR-429 and DDIT4 in the serum of EC patients are associated with positive lymph node metastasis and infiltration degree≥1/2 cm(P<0.05).The survival rate of patients with low serum miR-429 expression(36.67%)was significantly lower than that of patients with high miR-429 expression(89.06%)(P<0.05),while the survival rate of patients with high DDIT4 expression(35.48%)was significantly lower than that of patients with low DDIT4 expression(91.94%)(P<0.05).The degree of muscle wall infiltration≥1/2 cm,positive lymph node metastasis,low expression of miR-429,and high expression of DDIT4 were influencing factors for th
