摘 要:目的 研究红景天苷联合络塞维对缺血性脑卒中大鼠的作用及机制。方法 线栓法制备MCAO大鼠,分为假手术组,模型对照组,红景天苷联合络塞维组,阳性药对照组,给药7 d。测脑梗死体积及神经功能评分,RT-qPCR/ELISA测IL-1β、TNF-α、IL-6水平,免疫荧光染色/Western blot测NeuN、BDNF、TGF-β1、p-Smad蛋白;侧脑室注射TGF-β受体抑制剂,30 min后MCAO造模,给药7 d,检测以上指标。OGD/R诱导原代神经细胞,分为正常对照组、模型对照组、红景天苷联合络塞维组、TGF-β受体抑制剂+联合给药组,给药24 h,检测以上指标。结果 红景天苷联合络塞维能明显降低MCAO大鼠缺血侧脑梗死体积,改善神经功能,促进NeuN、BDNF、TGF-β1、p-Smad蛋白表达,抑制IL-1β、TNF-α及IL-6水平,且以上作用被TGF-β受体抑制剂阻断。同时,红景天苷联合络塞维能促进OGD/R诱导的原代神经细胞NeuN、BDNF蛋白,抑制IL-1β、TNF-α、IL-6,与体内结果相吻合。结论 红景天苷联合络塞维对MCAO大鼠及原代神经细胞具有保护作用,其作用机制与调控TGF-β信号通路密切相关。Aim To study the effect of salidroside combined with rosavin on ischemic stroke in rats.Methods The model of MCAO was established by using thread-embolic method.The rats were divided into the sham group,MCAO group,salidroside combined with rosavin group,positive control group,and the drug was given continuously for seven days.The infarct volume was measured by MRI and neurological deficit score was evaluated by Zea-Longa.The levels of NeuN,BDNF,TGF-β1,p-Smad were observed by Western blot and immunofluorescence staining.The expressions of IL-1β,TNF-αand IL-6 were performed by RT-qPCR/ELISA.The primary cortical neurons were isolated,OGD/R inducted,divided into the normal group,OGD/R group,salidroside combined with rosavin group,and TGF-β1 inhibitor+salidroside combined with rosavin group,the drug was given for 24 hours,and the expressions of NeuN,BDNF,IL-1β,TNF-αand IL-6 were measured.Results Salidroside combined with rosavin could decrease the infarct volume,improve the neurological function,promote the levels of Neun,BDNF,TGF-β1,p-Smad,and inhibit the expressions of IL-1β,TNF-αand IL-6.Salidroside combined with rosavin could promote NeuN,BDNF,inhibit IL-1β,TNF-α,IL-6 in primary nerve cells induced by OGD/R,and these effects were blocked by TGF-β1 inhibitor.Conclusions Salidroside combined with rosavin has neuroprotective effects on MCAO rats,and primary neurons are induced by OGD/R,and these effects are closely related to the TGF-βpathway.
