摘  要:目的:研究过表达lncRNA H19和miR-675及低表达miR-4500通过调控上皮间质转化和自噬促进乳腺癌细胞增殖、侵袭的机制及临床意义。方法:体外培养的乳腺癌MCF-7细胞分为过表达组、阴性对照组、空白组,每组设5个复孔。构建lncRNA H19过表达质粒,并以pc DNA3.1空白载体作为阴性对照,通过Lipofectamine 2000转染细胞,分别转染过表达组、阴性对照组的乳腺癌MCF-7细胞,检测对比各组癌细胞的增殖及侵袭能力,各组lncRNA H19和miR-675、miR-4500的mRNA相对表达量,各组N-cadherin、Vimentin、OPN、Snail、Slug等EMT相关基因的mRNA相对表达量及蛋白表达水平,以及各组LC3、Beclin1及mTOR蛋白表达水平。结果:过表达组的癌细胞增殖及侵袭能力较阴性对照组和空白组更强(P<0.05)。过表达组lncRNA H19和miR-675的mRNA相对表达量较阴性对照组和空白组更高(P<0.05),miR-4500的mRNA相对表达量较阴性对照组和空白组更低(P<0.05)。过表达组N-cadherin、Vimentin、OPN、Snail及Slug的mRNA相对表达量和蛋白表达水平较阴性对照组和空白组更高(P<0.05)。过表达组LC3及Beclin1的蛋白表达水平较阴性对照组和空白组更低,mTOR的蛋白表达水平较阴性对照组和空白组更高(P<0.05)。结论:过表达lncRNA H19和miR-675及低表达miR-4500能够调控上皮间质转化和自噬作用,进而促进乳腺癌细胞的增殖和侵袭,临床意义较大,值得关注。Objective:To study the mechanism and clinical significance of overexpression of lncRNA H19 and miR-675,lower expression miR-4500 in promoting breast cancer cell proliferation and invasion by regulating epithelial stromal transformation and autophagy.Methods:MCF-7 cells were divided into overexpression group,negative control group and blank group,with 5 complex wells in each group.The lncRNA H19 overexpression plasmid was constructed and the pc DNA3.1 blank vector as a negative control by Lipofectamine 2000 transfected cells with breast cancer MCF-7 cells from the overexpression and negative control groups,the proliferation and invasion capacity of each group of cancer cells,the relative mRNA expression of lncRNA H19 and miR-675,miR-4500 in each group,the relative mRNA expression and protein expression level of EMT-related genes including N-cadherin,Vimentin,OPN,Snail and Slug,as well as the protein expression levels of LC3,Beclin1 and mTOR proteins in each group were compared.Results:The proliferation and invasion ability of cancer cells in overexpression group was stronger than that of negative control group and blank group(P<0.05).The relative mRNA expression level of lncRNA H19 and miR-675 was higher in the overexpression group compared with the negative control group and blank group(P<0.05).The relative mRNA expression of miR-4500 was lower than that in the negative control group and blank group(P<0.05).The relative mRNA expression levels and protein expression levels of N-cadherin,Vimentin,OPN,Snail and Slug were higher in the overexpression group compared with the negative control group and blank group(P<0.05).The protein expression levels of LC3 and Beclin1 were lower than the negative control group and blank group,and the protein expression levels of mTOR was higher than the negative control group and the blank group(P<0.05).Conclusion:Overexpression of lncRNA H19 and miR-675,lower expression miR-4500 can regulate epithelial mal transition and autophagy,and then promote the proliferation and invasion of b
