天麻共生蜜环菌的液体发酵条件优化

摘  要:为实现天麻共生蜜环菌菌丝体的快速培养,采用液体摇瓶发酵的方法,以菌丝体生物量为主要评价指标,通过单因素试验筛选适宜的碳、氮源和松针浸出液浓度,采用正交试验设计分析得到三者添加到液体发酵培养基中的最优组合,对液体发酵培养基配方进行优化;以转速、装液量和接种量为变量,分别进行单因素试验并设计正交试验实现液体发酵条件的优化。结果表明:蜜环菌液体发酵培养中,最佳碳源为葡萄糖+木薯粉(1∶1),最佳氮源为酵母浸粉,松针浸出液的最佳浓度为1.5%;正交试验优化确定的液体发酵最佳培养基配方为葡萄糖10 g/L、木薯粉10 g/L、酵母浸粉12 g/L、松针15 g/L、磷酸二氢钾1.5 g/L、硫酸镁0.75 g/L,pH自然,最佳发酵条件为转速160 r/min,500 mL规格锥形瓶中装液量300 mL,接种量5.0%(V/V),25℃振荡培养7 d,在该条件下,得到的菌丝体生物量为12.42 g/L。In order to realize the rapid cultivation of the mycelium of Armillaria mellea symbiotic with Gastrodia elata,the method of liquid fermentation in shake-flask was used,with mycelial biomass serving as the primary evaluation metric.Suitable carbon and nitrogen sources,as well as the concentration of pine needle extract,were screened through single-factor experiments.The orthogonal experimental design was employed to analyze and determine the optimal combination of these three components for incorporation into the liquid fermentation medium,thereby optimizing the formulation of liquid medium.Subsequently,single-factor experiments were conducted with stirring speed,liquid volume,and amount of inoculum as variables.The orthogonal experimental design was also applied to these variables to optimize the liquid fermentation conditions.The results indicated that,during the liquid fermentation cultivation of A.mellea,the optimal carbon source was a combination of glucose and cassava starch(1:1),the best nitrogen source was the yeast extract,and the optimal concentration of the organic growth additive,pine needle extract,was 1.5%.The optimal liquid fermentation medium formulation determined through orthogonal experimental optimization was as follows:10 g/L glucose,10 g/L cassava starch,12 g/L yeast extract,15 g/L pine needles,1.5 g/L potassium dihydrogen phosphate,and 0.75 g/L magnesium sulfate,with a natural pH.The best fermentation conditions were a stirring speed of 160 rpm,a liquid volume of 300 mL in a 500 mL conical flask,the amount of inoculum was 5.0%(V/V),and incubation at 25℃with shaking for 7 days.Under these conditions,the obtained mycelial biomass was 12.42 g/L.
