摘 要:目的探索口腔鳞状细胞癌(OSCC)组织中脊髓灰质炎病毒受体(CD155)的表达及临床意义,探讨CD155对OSCC细胞迁移、增殖和侵袭的影响。方法使用免疫组织化学法(IHC)验证OSCC组织中CD155的表达,χ^(2)检验,COX回归分析CD155的表达与OSCC患者临床病理特征和预后的关系。通过蛋白免疫印迹(Western blot)和实时荧光定量PCR(RT-qPCR)验证CD155在OSCC细胞系中的转录水平。转染CD155干扰RNA后,通过体外细胞实验验证CD155对人舌鳞癌SCC15细胞的生物行为功能的影响。结果CD155在OSCC组织中主要表达于细胞膜,与癌旁正常组织相比表达显著升高(χ^(2)=50.750,P<0.0001);在Ⅲ+Ⅳ期(χ^(2)=25.488,P=0.001)、中低分化(χ^(2)=6.299,P=0.012)及T3+T4浸润深度(χ^(2)=23.820,P=0.001)、有淋巴结转移(χ^(2)=7.830,P=0.005)的患者中CD155的高表达率较高;高表达CD155与患者生存期较短有关(P<0.05);单因素和多因素COX回归分析显示:淋巴结转移是影响OSCC患者生存期较短的独立影响因素(P<0.05)。OSCC细胞中CD155蛋白表达及mRNA转录水平显著上调(P<0.05)。在转染siRNA-CD155的SCC15细胞中,实验组的增殖、迁移和侵袭能力显著低于对照组(P<0.05)。结论CD155在OSCC患者癌组织中表达较高,与患者预后不良相关;干扰CD155表达可对OSCC细胞生物学功能产生影响,抑制细胞增殖、迁移和侵袭的能力。Objective To investigate the expression and clinical significance of CD155 in oral squamous cell carcinoma(OSCC)tissues,as well as its impact on migration,proliferation and invasion.Methods Immunohistochemistry(IHC)was used to analyze the expression of CD155 in OSCC tissues and its correlation with clinicopathological features and prognosis.The transcription levels of CD155 were assessed by Western blot and RT-qPCR.Cell experiments were conducted to evaluate the effects of CD155 on OSCC cells following transfection with CD155 siRNA.Results CD155 was predominantly expressed on the cell membrane in OSCC tissues,with higher expression levels compared to normal tissues(χ^(2)=50.750,P<0.0001).High CD155 expression was associated withⅢ+IV disease stages(χ^(2)=25.488,P=0.001),poor differentiation(χ^(2)=6.299,P=0.012),T3+T4 depth of invasion(χ^(2)=23.820,P=0.001)and lymph node metastasis(χ^(2)=7.830,P=0.005)in OSCC patients;survival analysis revealed a correlation between high CD155 expression and shorter patient survival(P<0.05).COX regression analysis identified lymph node metastasis as an independent factor affecting the survival of OSCC patients(P<0.05).Both CD155 protein expression and mRNA transcription levels were significantly upregulated in OSCC cells(P<0.05).Transfection with siRNA-CD155 in OSCC cells resulted in significantly reduced proliferation,migration and invasion abilities compared to the control group(P<0.05).Conclusion CD155 is highly expressed in OSCC tissues and is associated with poor patient prognosis.Modulating CD155 expression can influence the biological functions of OSCC cells,leading to and inhibition of proliferation,migration,and invasion.
