摘 要:目的:优选竹节参多糖的最佳提取工艺,评价其体外抗氧化活性。方法:采用单因素试验和Box-Behnken响应面设计,以添加酶量、酶解温度、酶解时间、液料比作为考察因素,通过单因素实验得到最佳实验条件,利用Box-Behnken响应面法设计获得竹节参多糖的最佳酶法提取工艺;并采用1,1-二苯基-2-三硝基苯肼(DPPH)法测定竹节参多糖的抗氧化活性。结果:优化得到竹节参多糖酶法提取的最佳工艺为采用纤维素酶酶解;酶量为3%,酶解温度52℃,酶解时间2 h,液料比55 mL/g,此条件下预测多糖得率为86.19 mg/g,并具有较强的抗氧化活性。结论:优选出的竹节参多糖的提取工艺切实可行,竹节参多糖具有一定的抗氧化能力。Objective:Optimize the extraction process of polysaccharide from Panax japonicus and evaluate its antioxidant activity in vitro.Methods:Single factor design and Box-Behnken response surface design were used to optimize the best enzymatic extraction process of the polysaccharide from Panax japonicas with the added amount of enzyme,enzymatic hydrolysis temperature,enzymatic hydrolysis time,and solid-liquid ratio as factors.The antioxidant activity of polysaccharides was determined by DPPH method.Results:The optimum conditions for enzymatic extraction of polysaccharides from Panax japonicus were enzymatic hydrolysis for 2 h in 52℃water with liquid-to-material ratio of 55 mL/g by adding 3%cellulase.Under these conditions,the predicted yield of polysaccharides was 86.19 mg/g,and it had strong antioxidant activity.Conclusion:The extraction process of polysaccharides from Panax japonicas was feasible and the polysaccharides from Panax japonicas had antioxidant activity.
