摘 要:目的研究甲状腺激素T3对急性酒精性肝损伤小鼠的肝脏结构与功能的影响及细胞增殖与凋亡的相关基因表达。方法对正常小鼠提前给予不同浓度的甲状腺激素T3腹腔注射干预,最后一次给药后给予50%酒精溶液一次性灌胃,24 h后处死,并留取血清和肝脏。该实验通过建立小鼠急性酒精性肝损伤模型,对甲状腺激素T3在其中的作用进行分析。结果经T3干预后的急性酒精性肝损伤小鼠可观察到肝小叶轮廓清晰,肝索排列较为整齐,但存在炎细胞浸润,血清ALT活性和肝脏ROS含量增加,高剂量组PCNA表达增加(P<0.05),凋亡基因Caspase3和Bax表达较模型组有所降低(P<0.01),但随T3干预浓度升高凋亡基因表达出现上升趋势。结论浓度较高的甲状腺激素T3干预,会使小鼠急性酒精性肝损伤加重。Objective To study the effects of thyroid hormone T3 on the liver structure and function,as well as the expression of genes related to cell proliferation and apoptosis in mice with acute alcoholic liver injury.Methods Normal mice were intervened by intraperitoneal injection of different concentrations of thyroid hormone T3 in advance.After the last dose,50%alcohol solution was administered once by gavage.Twenty-four hours post-treatment,the mice were euthanized,and both serum and liver samples were collected.This experiment sought to establish a mouse model of acute alcoholic liver injury and assess the influence of thyroid hormone T3 on this condition.Results After T3 intervention,more severe damage in mice with acute alcoholic liver injury could be observed,clear liver lobule contours and neat arrangement of liver cords.But there was infiltration of inflammatory cells,increased serum ALT activity and liver ROS content,increased PCNA expression in the high-dose group(P<0.05),and decreased expression of apoptotic genes Caspase3 and Bax compared to model group(P<0.01).However,with the increase of T3 intervention concentration,the expression of apoptotic genes showed an upward trend.Conclusion Intervention with high concentrations of thyroid hormone T3 could exacerbate acute alcoholic liver injury in mice.
