摘 要:目的:观察核纤层蛋白B1(LMNB1)在胃癌组织中的表达,探究其与临床病理特征和预后的关系。方法:回顾性收集2017年1月至2021年11月山西省人民医院124例胃癌患者的癌组织及癌旁组织标本,通过集成高通量组织芯片,采用免疫组织化学染色法观察胃癌组织及其癌旁组织中LMNB1的表达情况,通过符号检验对比LMNB1在癌组织及癌旁组织中的表达差异。通过Mann-Whitney U分析明确LMNB1在不同胃癌病理资料中的表达差异,同时通过spearman等级相关性分析明确LMNB1表达与胃癌预后的关系,通过Kaplan-Meier生存分析明确LMNB1基因表达与胃癌相关生存期的联系。结果:使用高通量免疫组织化学染色检测人胃癌组织和癌旁组织LMNB1的表达,进行符号检验分析,发现LMNB1在胃癌组织中的表达高于癌旁组织[0.50(62/124)比0.03(5/135),Z=-9.589,P<0.05]。LMNB1的表达在不同性别中有显著差异[男性比女性为0.43(39/89)比0.65(23/35),R=0.197,P<0.05],与年龄[0.40(26/64)比0.60(36/60),R=0.194,P<0.05]、淋巴结转移数[0.40(25/61)比0.58(37/63),R=0.177,P<0.05]及人表皮生长因子受体2(Her-2)表达[0.46(39/83)比0.70(19/27),R=0.202,P<0.05]呈正相关,与血管内皮生长因子(VEGF)表达[0.63(29/46)比0.43(28/65),R=-0.197,P<0.05]呈负相关,同时LMNB1高表达患者总生存期[(36.050±24.818)个月比(51.043±25.980)个月,χ^(2)=6.075,P<0.05]和无病生存期[(36.130±24.385)个月比(51.150±25.781)个月,χ^(2)=6.117,P<0.05]显著低于LMNB1低表达患者,LMNB1高表达与患者生存期缩短显著相关。结论:胃癌组织中核纤层蛋白LMNB1呈高表达状态。LMNB1基因可能与胃癌的发生发展及预后明显相关。Objective:To clarify the expression of Lamin B1(LMNB1)in gastric cancer tissues,and explore the relationship between lamin B1 and clinicopathological features and prognosis.Methods:Cancer tissue and para-cancer tissue specimens of 124 patients with gastric cancer in Shanxi People’s Hospital January 2017 to November 2021 were retrospectively collected.The expression of LMNB1 in gastric cancer tissues and para-cancer tissues was observed by immunohistochemical staining with integrated high-throughput tissue chip.The expression difference of LMNB1 in cancer tissues and adjacent tissues was compared by symbol test.The expression difference of LMNB1 in different gastric cancer pathological data was determined by Mann-Whitney U analysis,and the relationship between LMNB1 expression and prognosis of gastric cancer was determined by spearman correlation analysis.Results:Using high-throughput immunohistochemical staining,the expression of LMNB1 in gastric cancer tissues and adjacent non-cancerous tissues was detected,and a sign test analysis was conducted.It was found that LMNB1 expression was higher in gastric cancer tissues than in adjacent non-cancerous tissues[0.50(62/124)vs.0.03(5/135),Z=-9.589,P<0.05].There was a significant difference in LMNB1 expression between genders[0.43(39/89)in males vs.0.65(23/35)in females,R=0.197,P<0.05],as well as in relation to age[0.40(26/64)in younger patients vs.0.60(36/60)in older patients,R=0.194,P<0.05],lymph node metastasis number[0.40(25/61)vs.0.58(37/63),R=0.177,P<0.05],and human epidermal growth factor receptor-2(Her-2)expression[0.46(39/83)vs.0.70(19/27),R=0.202,P<0.05],showing a positive correlation.LMNB1 expression also showed a negative correlation with vascular endothelial growth factor(VEGF)expression[0.63(29/46)vs.0.43(28/65),R=-0.197,P<0.05].Additionally,patients with high LMNB1 expression had significantly shorter overall survival[(36.050±24.818)months vs.(51.043±25.980)months,χ^(2)=6.075,P<0.05]and disease-free survival[(36.130±24.385)months vs.(51.150±25.781)m
