摘 要:以牛大力茎为研究对象,采用超声辅助乙醇法提取总黄酮。通过单因素实验考察了乙醇体积分数、液料比、提取时间、提取温度对牛大力茎总黄酮提取量的影响,并在此基础上,采用响应面法进一步优化总黄酮提取工艺。结果表明,在乙醇体积分数为70%、液料比为41∶1(mL∶g)、提取时间为42 min、提取温度为62℃的最优条件下,牛大力茎总黄酮提取量为16.06 mg·g^(-1),与预测值(16.23 mg·g^(-1))接近。该优化提取工艺操作简单,提取量高,为牛大力茎的利用及产品研发提供了依据。Taking Millettia speciosa Champ.stem as a research object,we extracted total flavonoids by ultrasonic-assisted ethanol extraction method.Moreover,we investigated the effects of ethanol volume fraction,liquid-solid ratio,extraction time,and extraction temperature on the extraction amount of total flavonoids from Millettia speciosa Champ.stem by single-factor experiments.Furthermore,we optimized the extraction process of total flavonoids by response surface methodologies.The results show that the extraction amount of total flavonoids from Millettia speciosa Champ.stem is 16.06 mg·g^(-1),which is close to the predicted value(16.23 mg·g^(-1))under the optimal conditions as follows:the ethanol volume fraction of 70%,the liquid-solid ratio of 41∶1(mL∶g),the extraction time of 42 min,and the extraction temperature of 62℃.The optimized extraction process is simple to operate,and has a high extraction amount,providing a basis for the utilization and product research and development of Millettia speciosa Champ.stem.
