摘  要:目的观察氢气对原代乳腺癌细胞的作用并探讨影响作用的因素,为氢气在乳腺癌临床治疗方面的应用提供实验依据。方法选取2022-02-10-2023-12-08临沂市妇幼保健院首次确诊的非特殊类型浸润性乳腺癌患者22例,取其手术切除的新鲜癌组织进行原代贴壁培养。原代乳腺癌细胞分为对照组和氢气组。对照组采用含10%血清的RPMI 1640培养液,氢气组采用含10%血清+饱和氢气的RPMI 1640培养液。观察癌细胞对氢气干预的反应,采用MTT检测细胞活性、酶联免疫吸附试验(ELISA)检测细胞凋亡。进一步探讨影响乳腺癌细胞对氢气反应性的因素,采用流式细胞仪检测乳腺癌细胞内活性氧(ROS)水平,收集患者临床及病理资料,分析细胞内ROS水平及患者临床病理指标与乳腺癌细胞对氢气反应性的关系。定量数据分析采用t检验,定性数据分析采用χ^(2)检验,P<0.05为差异有统计学意义。结果22例患者的乳腺癌细胞原代培养成功。其中14例氢气组(0.303±0.092)MTT光密度显著低于对照组(0.572±0.126),差异有统计学意义,t=2.925,P=0.004;ELISA吸光度氢气组(0.482±0.095)显著高于对照组(0.381±0.102),差异有统计学意义,t=2.379,P=0.032;说明氢气能够抑制癌细胞活性,增强癌细胞凋亡,干预有效。其余8例氢气组(0.512±0.062)MTT与对照组(0.533±0.093)比较,差异无统计学意义,t=0.963,P=0.353;氢气组(0.502±0.086)ELISA吸光度与对照组(0.491±0.096)比较,差异无统计学意义,t=0.832,P=0.395;说明氢气干预无效。比较14例与8例乳腺癌细胞内ROS水平及患者临床病理指标,前者(4.73±0.68)细胞内ROS水平显著高于后者(3.82±0.43),差异有统计学意义,t=3.015,P=0.003;前者(≥10%)病理组织切片免疫组织化学染色癌细胞Ki-67阳性率显著高于后者(<10%),差异有统计学意义,χ^(2)=7.056,P=0.004;前者(8例,57%)脉管内癌栓发生率显著高于后者(3例,38%),差异有统计学意义,χ^(2)=5.963,P=0.021Objective To observe the effect of hydrogen on primary breast cancer cells and explore the influencing factors,so as to provide experimental basis for the application of hydrogen in the clinical treatment of breast cancer.Methods A total of 22 patients with newly diagnosed non-specific invasive breast cancer from February 10,2022,to December 8,2023,were selected,and fresh cancer tissues from surgical resection were used for primary adherent culture.Primary breast cancer cells were divided into control group and hydrogen group.The control group was cultured in RPMI 1640medium containing 10%serum,while the hydrogen group was cultured in RPMI-1640 medium containing 10%serum and saturated hydrogen.The response of cancer cells to hydrogen gas intervention was observed.MTT assay was used to detect cell viability,and ELISA method was used to detect cell apoptosis.To further explore the factors that affect the responsiveness of breast cancer cells to hydrogen,flow cytometry was used to detect the level of reactive oxygen species(ROS)in breast cancer cells.Clinical and pathological data of patients were collected to analyze the relationship between the level of ROS in cells,clinical and pathological indicators of patients and the responsiveness of breast cancer cells to hydrogen.Quantitative data analysis was performed using t-test,while qualitative data analysis was performed using chi square test.A P-value less than 0.05 was considered statistically significant.Results The primary culture of breast cancer cells from 22 patients was successful.MTT absorbance of hydrogen group(0.303±0.092)of cancer cells from 14patients of was significantly lower than that of the control group(0.572±0.126),t=2.925,P=0.004,the difference was statistically significant;ELISA absorbance of hydrogen group(0.482±0.095)was significantly higher than that of the control group(0.381±0.102),t=2.379,P=0.032,and the difference was statistically significant;Hydrogen gas can inhibit cancer cell activity,enhance cancer cell apoptosis,and intervene ef
