摘  要:目的探讨恶性胸膜间皮瘤(MPM)实验模型的优缺点,总结不同实验模型在MPM基础研究中的使用。方法以“恶性胸膜间皮瘤、临床前模型、细胞株、动物模型”为中文关键词,“malignant pleural mesothelioma,preclinical models,cell lines,animal models”为英文关键词,检索PubMed及中国知网2019-01-01-2023-07-01相关文献。纳入标准:(1)MPM临床前模型相关研究;(2)MPM动物模型相关文献;(3)MPM细胞株模型相关文献。排除标准:MPM实验模型相关性较低及数据较陈旧文献。最终共纳入文献68篇。结果MPM因发病潜伏期长及症状的隐匿性,导致其诊疗困难,确诊时基本为晚期,需建立多种不同实验模型进行临床基础研究。本研究综述不同实验模型的分类、制作方法、优缺点及用途,分析临床研究中常用的体外细胞株和体内动物模型的优缺点,结果显示,体外细胞株模型(人类或动物MPM细胞)主要用于基础细胞实验,体内动物模型主要用于化疗免疫新制剂的研发实验。通过对目前使用的各种MPM临床前模型描述,加深对MPM生物学的理解,为基础实验使用临床前模型提供可靠依据,为探索MPM诊治提供新思路与方法,为提高MPM诊疗水平奠定了基础。结论分析体外细胞株和体内动物模型优缺点,得出模型产生的肿瘤与人类MPM特征有一定差距。但随着分子生物学和遗传学的发展,更多的MPM转基因鼠模型被开发,正逐渐减小此差距。目前各种模型均存在不足,现急需优化存在的劣势,从而获得更多不同用途的模型。Objective To discuss the advantages and disadvantages of different experimental models of malignant pleural mesothelioma(MPM)and summarize the appropriate use of different experimental models in MPM basic research.Methods With"malignant pleural mesothelioma,preclinical model,cell line,animal model"as the Chinese keywords,"malignant pleural mesothelioma,preclinical models,cell lines,animal models"as the English keywords,relevant literature on PubMed and CNKI were searched from January 1,2019 to July 1,2023.Inclusion criteria:(1)MPM preclinical model related research;(2)MPM animal model related literature;(3)MPM cell line model related literature.Exclusion criteria:literature with low relevance to MPM experimental models and outdated data.Finally 68 literatures were selected.Results The diagnosis and treatment of MPM are difficult due to the long incubation period of the disease and the concealment of symptoms,and the diagnosis is basically late,so it is necessary to establish a variety of different experimental models for clinical basic research.This study has analyzed the classification,production methods,advantages and disadvantages of different experimental models,analyzed the advantages and disadvantages of in vitro cell lines commonly used in clinical research and in vivo animal models,and concluded that in vitro cell line models(human or animal MPM cells)are mainly used for basic cell experiments,while in vivo animal models are mainly used for the research and development of new agents of chemotherapy and immunity.Through the description of various MPM preclinical models currently in use,we can deepen our understanding of MPM biology and provide a reliable basis for the use of preclinical models in basic experiments.It lays a foundation for exploring new ideas and methods of MPM diagnosis and treatment and improving the level of MPM diagnosis and treatment.Conclusions By analyzing the advantages and disadvantages of in vitro cell lines and in vivo animal models,it is concluded that there is a certain gap bet
