摘 要:为探讨益母草碱(leonurine,LEO)对过敏性紫癜肾炎(Henoch-Schonlein purpura nephritis,HSPN)的影响,将小鼠分为空白对照组、模型组、LEO低剂量[25 mg/(kg·d)]组、LEO中剂量[50 mg/(kg·d)]组、LEO高剂量[100 mg/(kg·d)]组,用考马斯亮蓝法检测24 h尿白蛋白(albumin,Alb)水平并计数尿红细胞;全自动生化分析仪检测血清肌酐(creatinine,Cr)、血尿素氮(blood urea nitrogen,BUN)水平;免疫荧光染色检测肾组织IgA沉积情况;H-E染色观察肾脏组织病理学变化;ELISA检测血清IL-4、IL-10、IFN-γ及TNF-α水平;聚乙二醇沉淀比浊法检测血清循环免疫复合物水平;流式细胞术测定外周血Th1/Th2比值。结果显示,与空白对照组比较,模型组小鼠24 h尿Alb水平,尿红细胞数,血清Cr、BUN、IL-4、IL-10、循环免疫复合物水平显著升高,血清IFN-γ、TNF-α水平及外周血Th1/Th2比值显著降低(P<0.05);与模型组比较,LEO低、中、高剂量组小鼠24 h尿Alb水平,尿红细胞数,血清Cr、BUN、IL-4、IL-10、循环免疫复合物水平显著降低,血清IFN-γ、TNF-α水平及外周血Th1/Th2比值显著升高(P<0.05);LEO作用效果呈剂量依赖性(P<0.05)。该研究提示,LEO可减轻HSPN小鼠肾组织损伤,改善肾功能,发挥肾保护作用,其作用机制可能与促进Th1/Th2平衡和促进机体免疫功能恢复正常有关。To investigate the effect of leonurine(LEO)on Henoch-Schonlein purpura nephritis(HSPN),mice were divided into the blank control group,the model group,the low(25 mg/[kg·d]),medium(50 mg/[kg·d])and high(100 mg/[kg·d])doses of LEO treatment groups.The 24-hour urinary albumin(Alb)content was measured by the Bradford method and urine red blood cells were counted.The serum creatinine(Cr)and blood urea nitrogen(BUN)were measured by the automatic biochemical analyzer.Immunofluorescence staining was used to detect IgA deposition in renal tissues.The histopathological changes in renal tissues were observed by H-E staining.ELISA was performed to detect serum IL-4,IL-10,IFN-γ,and TNF-α.Serum circulating immune complex levels were measured by polyethylene glycol precipitation turbidimetry,and the peripheral blood Th1/Th2 ratio was measured by flow cytometry.The results showed that compared to those of the blank control group,the 24-hour urinary Alb level,urine red blood cell count,and serum levels of Cr,BUN,IL-4,IL-10,and circulating immune complex of the model group were significantly higher,whereas the serum IFN-γ,TNF-αlevels,and peripheral blood Th1/Th2 ratio were significantly lower(P<0.05).Compared to those of the model group,the 24-hour urinary Alb level,urine red blood cell count,serum Cr,BUN,IL-4,IL-10,and circulating immune complex levels were significantly reduced,while the serum IFN-γ,TNF-αlevels,and peripheral blood Th1/Th2 ratio were significantly increased in all the LEO treatment mice(P<0.05).In addition,the effect of LEO was dose-dependent(P<0.05).Taken together,this study suggests that LEO treatment reduces renal tissue damage and improves renal function and renal protection in HSPN mice.The underlying mechanism may be related to the promotion of Th1/Th2 balance which helps to restore normal immune function.
