摘 要:文章研究了沉香叶黄酮对油酸诱导HepG2细胞脂质代谢的影响。采用水溶性四唑盐法检测细胞增殖率,利用试剂盒检测细胞内脂质生成相关因子水平,包括甘油三酯(Triglyceride,TG)、总胆固醇(Total cholesterol,TC)、低密度脂蛋白胆固醇(Low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(High-density lipoprotein cholesterol,HDL-C);同时检测氧化应激相关因子水平,如活性氧(Reactive oxygen species,ROS)、丙二醛(Malondialdehyde,MDA)、超氧化物歧化酶(Superoxide dismutase,SOD)、谷胱甘肽(Glutathione,GSH)、过氧化氢酶(Catalase,CAT)水平。结果表明:沉香叶黄酮可以减少油酸诱导的Hep G2细胞内TG、TC、LDL-C、MDA、ROS释放水平,促进HDL-C释放,提高SOD、GSH、CAT活性。沉香叶黄酮可以缓解油酸诱导的HepG2细胞脂质代谢紊乱,其作用可能与调节细胞脂质代谢、清除活性氧、提高抗氧化酶活性有关。The study investigated the impact of flavonoids from agarwood leaves on lipid metabolism in HepG2 cells induced by oleic acid.Cell proliferation was assessed using the water-soluble tetrazolium salt method.Biochemical kits were employed to measure lipid metabolism-related factors levels within cells,including triglycerides(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C).Additionally,oxidative stress-related factors levels such as reactive oxygen species(ROS),malondialdehyde(MDA),superoxide dismutase(SOD),glutathione(GSH),and catalase(CAT)were measured.The results showed that flavonoids from agarwood leaves reduced the levels of TG,TC,LDL-C,MDA,and ROS released in oleic acid-induced HepG2 cells,promoted the release of HDL-C,and increased the activities of SOD,GSH,and CAT.Agarwood leaf flavonoids can alleviate oleic acid-induced lipid metabolism disorders in HepG2 cells,its effect may be related to the regulation of cellular lipid metabolism,scavenging of reactive oxygen species,and enhancement of antioxidant enzyme activity.
