SHP-1基因在卵巢癌中的表达及其与患者预后关系生物信息学分析

摘  要:目的 采用生物信息分析方法探讨SHP-1基因在卵巢癌中的表达、信号通路和预后关系.方法 在癌症基因组图谱(TCGA)数据库中检索SHP-1基因,分析SHP-1基因在人实体肿瘤中的表达情况.应用相互作用基因/蛋白在线检索工具(STRING)建立SHP-1基因相互作用蛋白网络,并对网络中的基因进行本体论和KEGG信号通路富集.依据SHP-1基因在卵巢癌组织中的差异表达情况分为高、低表达组,对比高表达与低表达患者总生存(OS)和无疾病进展生存(DFS)是否存在差异.结果 在人体不同肿瘤组织中SHP-1基因mRNA表达水平存在一定的差异,在泌尿生殖系统肿瘤、睾丸癌和肺癌中表达水平较高,而在胶质瘤、前列腺癌和恶性色素瘤中表达水平较低.PPI网络中相关蛋白21个,相互作用连接118个,蛋白间平均关联度为11.2,区域聚类指数为0.807,蛋白网络富集显著(P<0.001).SHP-1基因相关信号通路主要富集于EGFR相关信号通路、Tp53信号通路、Jak-STAT信号通路等.SHP-1基因主要突变形式为无意义突变、缺失突变和同义突变等.SHP-1基因高表达组患者OS(HR=0.95)和DFS(HR=0.85)均显著高于低表达组,但差异尚无统计学意义(P>0.05).结论 SHP-1基因与卵巢癌患者的预后有关,高表达者生存期延长,可能成为卵巢癌患者预后较好的分子标志物.Objective To explore the expression,signaling pathway,and prognostic relationship of SHP-1 gene in ovarian cancer using bioinformatics analysis methods.Methods Retrieve SHP-1 expression model was evaluated in TCGA and analyze its expression in human solid tumors.Establish a SHP-1 gene interacting protein network using the STRING online retrieval tool,and enrich the genes in the network through ontology and KEGG signaling pathways.According to the differential expression of SHP-1 gene in ovarian cancer tissues,it is divided into high and low expression groups,and compared in OS and DFS.Results There are certain differences in the mRNA expression level of SHP-1 gene in different tumor tissues of the human body.The expression level is higher in urinary system tumors,testicular cancer,and lung cancer,while it is lower in glioma,prostate cancer,malignant and melanoma.There are 21 related proteins in the PPI network,with 118 interacting connections.The average correlation between proteins is 11.2,and the regional clustering index is 0.807.The protein network is significantly enriched(P<0.001).The SHP-1 gene related signaling pathway is mainly enriched in EGFR related signaling pathway,Tp53 signaling pathway,Jak STAT signaling pathway,etc.The main mutation forms of SHP-1 gene are meaningless mutations,deletion mutations,and synonymous mutations.The OS(HR=0.95)and DFS(HR=0.85)in SHP-1 high expression group were higher than low group,but without statistical difference(P>0.05).Conclusion The SHP-1 gene is associated with the prognosis of ovarian cancer patients,with high expression significantly prolonging survival and potentially becoming a molecular marker for better prognosis in ovarian cancer patients.
