AURKB表达与结直肠癌临床病理特征及药物敏感性相关性分析

摘　　要：目的 为改善结直肠癌患者预后,寻找结直肠癌治疗敏感性靶点。方法 通过免疫组化染色方法检测155例结直肠癌术后患者病理组织。利用Kaplan-Meier单因素生存、单因素COX回归分析Aurora激酶B(Recombinant Aurora Kinase B,AURKB)表达与结直肠癌患者预后的关系;通过配对t检验分析AURKB在不同分类标准下癌组织与正常组织的差异表达;最后利用在线数据可以分析AURKB表达与临床特征、药物敏感性信号通路及药物敏感性相关性。结果 AURKB蛋白在结直肠癌中低表达;AURKB蛋白表达与结直肠癌患者预后无相关性;AURKB表达与G2M期、DNA修复具有中度强度正相关性;AURKB高表达有利于靶向药AKT抑制剂(A.443654)、阿法替尼(BIBW2992)、阿西替尼(Axitibinb)、顺铂(Cisplatin)、阿糖胞苷(Cytarabine)、RSK抑制剂(SL.0101.1)、极光激酶抑制剂(ZM.447439)药物治疗结直肠癌患者,AURKB低表达有利于RAF抑制剂(AZ6281)、BRAF抑制剂(PLX4720)药物治疗结直肠癌患者。结论 AURKB可以作为结直肠癌患者药物治疗敏感性的靶标。Objective To improve the prognosis of patients with colorectal cancer(CRC) and identify targets for treatment sensitivity.Methods Immunohistochemical staining was conducted on pathological tissues from 155 CRC patients post-surgery.The relationship between Aurora kinase B(AURKB) expression and patient prognosis was analyzed using Kaplan-Meier univariate survival and univariate COX regression.The expression of AURKB in cancer and normal tissues was analyzed by paired T-test.Finally,online data analysis was used to explore the correlation between AURKB expression,clinical characteristics and drug sensitivity.Results AURKB expression was found to be low in CRC but there was no significant correlation between its expression and CRC patient prognosis.AURKB expression had moderate positive correlation with G2M stage and DNA repair processes.High AURKB expression was associated with improved treatment outcomes for CRC patients using AKT inhibitors(A.443654),afatinib(BIBW2992),axitinib,cisplatin,cytarabine,RSK inhibitors(SL.0101.1),and aurora kinase inhibitors(ZM.447439).Conversely,low AURKB expression was beneficial for treatment with RAF inhibitors(AZ6281) and BRAF inhibitors(PLX4720)Conclusion AURKB can be used as a target for drug sensitivity in patients with CRC.
