酸枣仁、龙眼肉提取物复合酪蛋白肽和GABA组方对ICR小鼠睡眠改善作用的研究

摘  要:本文以龙眼肉提取物(58.1%)和酸枣仁提取物(24%)为主要成分,添加酪蛋白水解物(10.9%)和γ-氨基丁酸(γ-aminobutyric acid,GABA,7%)制备复合功能食品SLγL,研究其对ICR小鼠的睡眠改善作用及其功效的分子机制研究。研究结果表明,SLγL高中低剂量组均显著增加戊巴比妥钠诱导小鼠的睡眠时间,并降低巴比妥钠睡眠潜伏期。SLγL高中剂量组可显著提高小鼠睡眠发生率(P <0.05)。SLγL组方各剂量组可上调小鼠海马和下丘脑组织中的5-HT1AR、GABAARα1和GABAARγ2蛋白的表达水平,以及小鼠海马NKCC1蛋白的表达水平(P <0.05)。SLγL组方各剂量组对小鼠的体重和直接睡眠均无显著(P>0.05)影响,对小鼠海马和下丘脑组织中5-HT和GABA的水平的影响也均无统计学意义(P> 0.05)。综上所述,SLγL组方可能通过上调小鼠海马和下丘脑组织中5-HT和GABA受体蛋白的表达改善ICR小鼠的睡眠。本研究可为开发改善睡眠质量的功能食品提供理论和数据支持。In this paper,Semen ziziphi spinosae(SZS)extracts(24%)and longan pulp extracts(58%)were used as the main ingredients,and casein hydrolysate(11%)andγ-aminobutyric acid(GABA)(7%)were added to prepare a functional food compound named SLγL,its sleep-improving effects on ICR mice and its molecular mechanism were studied.The results showed that all dosage groups of SLγL significantly increased the sleep duration and decreased sleep latency of pentobarbital sodium-induced mice,meanwhile,the high-dose and medium-dose groups of SLγL significantly increased the sleep incidence rate of pentobarbital sodium-induced mice(P<0.05).all dosage groups of SLγL up-regulated the expression levels of 5-HT1AR,GABAARα1 and GABAARγ2 proteins in the hippocampus and hypothalamus of mice,as well as mice hippocampal NKCC1 protein expression level(P<0.05).SLγL had no significant effect on the body weight and direct sleep of mice(P>0.05),and the effects on the levels of 5-HT and GABA in mice hippocampal and hypothalamic tissues were not statistically significant(P>0.05).In conclusion,the SLγL compound might improve sleep in ICR mice by up-regulating the expression of 5-HT and GABA receptor proteins in mice hippocampal and hypothalamic tissues.This study provides theoretical and data support for the development of functional foods aimed at improving sleep quality.
