摘  要:采用甲基绿吸收光谱法同时测定糖果中酒石黄和日落黄含量,并对不同糖果前处理及甲基绿吸收光谱法检测条件进行单因素优化。结果表明,不同糖果前处理条件略有差异。在400~800 nm范围内,Tris-HCl用量1 m L,p H为9.43,甲基绿溶液用量2 m L,反应时间15 min时,甲基绿与酒石黄和日落黄发生显色反应生成离子缔合物,酒石黄-甲基绿在574 nm处测得最大值,标准曲线为y=-0.0035x+0.7009(R^(2)=0.9993),线性范围是0.03~16 mg/L,检出限为0.069 mg/L;日落黄-甲基绿在488 nm处测得最大值,标准曲线为y=0.0378x+0.2104(R^(2)=0.9993),线性范围是0.04~14 mg/L,检出限为0.024 mg/L。对实际样品进行加标回收,酒石黄与日落黄的加标检测均在正常范围内。该方法简便、快速、灵敏度高,可以在不同波长下同时测定硬质糖果中的酒石黄和日落黄的含量。Using methyl green absorption spectroscopy to simultaneously determine the content of tartar yellow and sunset yellow in candies,various sample pretreatment conditions for different types of candies and detection parameters for the methyl green absorption spectroscopy were optimized through single-factor experiments.The results indicated that there were slight differences in pre-treatment conditions for different candies.In the wavelength range of 400〜800 nm,optimal conditions were found with 1 mL of Tris-HCl buffer at pH 9.43,2 mL of methyl green solution,and a reaction time of 15 minutes.Under these conditions,methyl green formed ion associates with both tartar yellow and sunset yellow,yielding maximum absorbance at 574 nm for the sunset yellow methyl green complex,with a standard curve of y=-0.0035x+0.7009(R^(2)=0.9993),with a linear range of 0.03 to 16 mg/L,and a detection limit of 0.069 mg/L.The maximum value of sunset yellow methyl green complex was measured at 488 nm,with a standard curve of y=0.0378x+0.2104(R^(2)=0.9993),a linear range of 0.04〜14 mg/L,and a detection limit of 0.024 mg/L.Spiked recovery tests on actual samples confirmed that the detection of both tartar yellow and sunset yellow was within normal limits.This method was simple,fast,highly sensitive,and had good analytical results.It can simultaneously determine the content of tartar yellow and sunset yellow in hard candies at different wavelengths.
