摘 要:随着生物信息学和公共数据库的不断发展,越来越多的癌症相关基因被发现。突触结合蛋白(synaptotagmins,SYTs)是一类在突触小泡和嗜铬细胞颗粒中丰富存在的膜蛋白,其进化高度保守。由一个短的N-末端序列、一个可变连接结构域、一个跨膜结构域和两个C2结构域组成。SYTs在哺乳动物细胞中已被发现存在17种亚型,它们分布广泛且功能复杂。有研究表明SYTs在调节钙依赖的膜融合进程中发挥重要作用,一些家族成员还与激素分泌或神经递质释放相关。此外,有研究结果证实SYTs可以作为胞吐作用的钙离子感受器。近年来,大量研究表明SYTs与多种肿瘤的发生发展高度相关。下调SYTs的表达抑制肿瘤细胞的增殖,迁移和侵袭,促进细胞凋亡。腹腔注射siSYT的小鼠腹膜结节的生长受到抑制,存活时间延长。因此,SYTs被认为是一类致癌基因,其有望成为多种癌症的预后生物标志物。该文概述了最近报道的关于SYTs家族成员在癌症中作用的研究,并突出已经取得的进展。More tumor-related genes were discovered with the development of bioinformatics and public databases.Synaptotagmins(SYTs)are a kind of membrane proteins abundant in synaptic vesicles and chromaffin cell granules,and their evolution is highly conserved.It consists of a short N-terminal sequence,a variable connectivity domain,a transmembrane domain,and two C2 domains.Seventeen subtypes of SYTs have been found in mammalian cells,which are widely distributed and have complex functions.Studies have shown that SYTs regulate calcium dependent membrane fusion events,and some family members are also associated with hormone secretion or neurotransmitter release.Many pieces of evidence support SYTs as a Ca^(2+)receptor of exocytosis.In recent years,it has been shown that SYTs are highly correlated with the occurrence and development of various tumors.Downregulating the expression of SYTs inhibits the proliferation,migration,and invasion,but promotes cell apoptosis of tumor cells.The growth of peritoneal nodules in mice injected with siSYT was inhibited and the survival time of the mice was prolonged.Therefore,SYTs can serve as a carcinogenic gene and potentially become a prognostic biomarker for various cancers.This article provides an overview of recent research on the role of SYT family members in cancer and highlights the progress that has been made.
