摘　　要：目的研究伊红组织固定液联合锥形底标本瓶在改善宫颈管搔刮术(ECC)制片中的应用。方法用含伊红固定液的锥形底标本瓶留取ECC标本,取材前标本瓶直接4000 r/min离心10 min,离心半径20 cm,弃上清,用镊子或长柄耳勺刮取全部组织,按取材规范包裹好,常规制片。结果经高速离心处理后,锥形底标本瓶底部可见沉底或附壁红色小凝块,易于取材;经常规制片后,镜下组织或细胞集中,无退变或收缩现象,染色清晰,对比度好,镜下能观察到颈管黏液腺上皮细胞、炎细胞,有时可见完整颈管黏膜组织,对免疫组织化学结果亦无影响,可明确判断宫颈管上皮细胞和病变的形态,能够满足病理诊断。结论利用含伊红组织固定液的锥形底标本瓶处理ECC标本,能够最大限度留取标本中的组织和细胞,此方法方便实用,效果良好。Objective To explore the application of a conical-bottomed specimen bottle containing eosin fixative in improving endocervical curettage(ECC)preparation.Methods We used a conical-bottomed specimen bottle containing eosin fixative to collect ECC specimens.Before sampling,the specimen bottle was directly centrifuged at 4000 r/min for 10 min.The supernatant was discarded,and all tissues were scraped with tweezers or long-handled ear spoons.ECC specimens were prepared according to the standard protocol.Results After high-speed centrifugation,small red clots could be seen at the bottom of the conical-bottomed specimen bottle or the wall,which made it easy to get the specimen.After routine preparation,the tissue or cells were concentrated under the microscope,without degeneration or shrinkage.The staining was clear with good contrast.Cervical mucous gland epithelial cells,inflammatory cells,and intact cervical canal mucosa could be observed under the microscope.It had no effect on the results of immunohistochemistry.It was easy to determine the morphology of cervical epithelial cells and lesions,which met pathological diagnosis.Conclusion The use of conical-bottom specimen bottles containing eosin fixative to prepare ECC specimens can maximize the retention of tissues and cells in the specimens.This method is convenient and practical,and the effect is good.
