摘 要:依赖自噬相关基因调控的自噬过程通过促进细胞内损伤老化物质降解和回收再利用对细胞生存及胞内稳态维持发挥至关重要的作用。核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎症小体作为固有免疫的主要组成成分在机体多种疾病发生及发展过程中发挥作用。自噬-NLRP3炎症小体通路与炎症性肠病、放射性肠炎、感染性肠炎及缺血性肠炎等发生和发展密切相关,主要通过降解NLRP3炎症小体组分、抑制活性氧产生等多种途径抑制肠道组织中NLRP3炎症小体激活,从而减轻肠道相关炎症反应。本文综述近5年关于自噬-NLRP3炎症小体通路在肠炎中作用和机制的研究进展及当前研究中发现的典型NLRP3炎症小体抑制剂,以期为肠炎相关新药研制提供新思路。The autophagy process regulated by autophagy-related genes plays a vital role in cell survival and maintenance of intracellular homeostasis by promoting degradation and recycling of damaged and aged substances in cells.As a major component of innate immunity,nucleotide-binding oligomeri zation domain-like receptors protein 3(NLRP3)inflammasome plays a role in the occurrence and development of various diseases in the body.The autophagy-NLRP3 inflammasome pathway is closely related to the occurrence and development of inflammatory bowel disease,radiation enteritis,infectious enteritis and ischemic enteritis.It mainly inhibits activation of NLRP3 inflammasome in intestinal tissue through various pathways such as degradation of NLRP3 inflammasome components and inhibition of reactive oxygen species production,thereby reducing intestinal related inflammatory reactions.This article reviews research progress on the role and mechanism of autophagy-NLRP3 inflammasome pathway in enteritis in the past five years,as well as typical NLRP3 inflammasome inhibitors discovered in current research,in order to provide new ideas for the development of new drugs related to enteritis.
