摘  要:目的探究肿瘤坏死因子-α诱导蛋白8样蛋白2(TIPE2)在不同程度牙周炎牙周组织中的表达及TIPE2在牙周炎发病过程中的作用机制。方法1)选取12例进行牙周手术的牙周炎志愿患者,排除患有糖尿病等系统性疾病史,根据牙周炎的严重程度将志愿患者分为重度组(n=6)和轻中度组(n=6),将牙周手术术中切除的牙龈组织进行免疫组织化学染色分析,观察TIPE2在不同程度牙周炎牙龈组织中的表达情况;2)体外培养人牙周膜干细胞(PDLSC),使用1μg·mL^(-1)脂多糖(Pg-LPS)模拟牙周炎炎症环境,在炎症微环境下将细胞分别培养24 h、48 h、72 h,通过细胞免疫组织化学染色及RT-qPCR检测,分析炎症微环境下PDLSC中TIPE2的基因表达水平变化。结果1)重度组牙龈组织中浸润的炎性细胞TIPE2阳性表达明显多于轻中度组(P<0.05);2)免疫组织化学染色提示,TIPE2在体外培养的PDLSC中呈阳性表达;3)将PDLSC分别在含1μg·mL^(-1) Pg-LPS的培养液中分别培养24 h、48 h、72 h后,RT-qPCR检测细胞中TIPE2基因表达水平,结果显示,细胞中的TIPE2基因表达量较空白对照组表达量均有所降低(P均<0.05)。结论TIPE2参与人牙周炎的疾病过程,且与炎症的严重程度密切相关,TIPE2可能在牙周炎炎症反应中发挥负调控作用。Objective This study aims to explore the expression of TIPE2 in periodontal tissues with different degrees of eriodontitis and development mechanism of periodontitis.Methods Firstly,12 patients with periodontitis who underwent periodontal surgery were selected,and patients with a history of systemic diseases were excluded.According to the severity of periodontitis,the volunteers were divided into severe group(n=6)and mild-moderate group(n=6).Then,immunohistochemical staining was performed on the gingival tissues excised during surgery to observe the expression of TIPE2 in gingival tissues with different degrees of periodontitis.Next,human periodontal ligament cells(PDLSC)were cultured in vitro and stimulated with 1μg·mL^(-1) Pg-LPS to simulate the inflammatory environment.Immunohistochemical staining and RT-qPCR were performed to detect the gene expression level of TIPE2 in PDLSC under inflammatory conditions.Results The number of TIPE2-positive inflammatory cells infiltrating in severe group gingival tissues was significantly higher than that in mild-moderate group(P<0.05);Immunohistochemical staining indicated positive expression of TIPE2 in PDLSC cultured in vitro;After stimulating PDLSC with Pg-LPS in the inflammatory microenvironment for 24 h,48 h,72 h,RT-qPCR showed that the gene expression level of TIPE2 in cells decreased compared with that in control group PDLSC.Conclusion TIPE2 is involved in the pathogenesis of periodontitis in humans and is closely related to the severity of inflammation.The gene expression level of TIPE2 in PDLSC decreases in the inflammatory microenvironment.The mechanism of action of TIPE2 in the progression of periodontitis needs further exploration.
