摘  要:芹菜是伞形科芹属一二年生草本植物,富含芹菜素和木犀草素为主的类黄酮物质。赤霉素是一种常见的植物激素,能够促进植物根茎快速延伸,并影响类黄酮物质积累。目前关于外施赤霉素对芹菜类黄酮物质积累的研究尚不深入。以黄芹品种‘黄太极’为研究材料,在播种60 d(60 DAS)后对叶面喷施120mg·L^(-1)的GA3溶液,5 d为一周期,处理两次。在三个时期(60、65、70 DAS),分别测定芹菜小叶片和叶柄中芹菜素及木犀草素的含量,并分析芹菜素与木犀草素合成关键酶基因(AgPAL、AgC4H、AgCHS、AgCHI、AgFNS)表达水平。结果表明,赤霉素处理可提高65、70 DAS小叶片中AgPAL、AgC4H、AgCHS、AgFNS的相对表达水平,促进两个阶段(60~65 DAS、65~70 DAS)木犀草素和第一阶段(60~65 DAS)芹菜素的积累;提高第一阶段叶柄中AgPAL、AgC4H、AgCHS、AgCHI、AgFNS的表达水平,促进芹菜素和木犀草素的积累;降低第二阶段(65~70 DAS)叶柄中AgPAL、AgC4H、AgFNS的表达水平,促进木犀草素的积累,对芹菜素含量无显著影响。本研究初步探索赤霉素影响芹菜类黄酮合成的内在机理,为深入研究外施赤霉素提高芹菜类黄酮含量提供了思路。Celery is a biennial herb of the genus apigenin in the Apiaceae family.It is rich in flavonoids mainly composed of apigenin and luteolin.Gibberellin(GA)is a plant hormone that can promote the extension of plant roots and affect the accumulation of flavonoids.At present,the effects of exogenous gibberellin on the accumulation of flavonoids in celery remain unclear.The 60 days after sowing(DAs)plants of yellow celery cultivar‘Huangtaij'were used as the research material.The 120 mg·L^(-1) GA,solution was sprayed on the foliar surface for 5 days as a cycle,and the treatments were twice.The contents of apigenin and luteolin in celery small leaves and petioles and the expression levels of key enzyme genes related to apigenin and luteolin synthesis(AgPAL,AgC4H,AgCHS,AgCHI,and AgFNS)were measured at three periods(60,65,and 70 DAS).The results showed that exogenous gibberellin treatment could increase the relative expression levels of AgPAL,AgC4H,AgCHS,and AgFNS in small leaves at two periods(65 and 70 DAS).It could promote the accumulation of luteolin in the two stages(60-65 DAS and 65-70 DAS)and apigenin in the first stage(60-65 DAS).The expression levels of AgPAL,AgC4H,AgCHS,AgCHI,and AgFNS in petioles were increased in the first stage(60-65 DAS).The accumulations of apigenin and luteolin were also promoted.The expression levels of AgPAL,AgC4H,and AgFNS in the petioles of the second stage(65-70 DAS)were decreased.The accumulation of luteolin had no significant effect on the content of apigenin in the stage.This study preliminarily explored the internal potential mechanism of gibberellin affecting the synthesis of flavonoids in celery.The results provided an idea for further study of the effect of gibberellin on the content of flavonoids in celery.
