枳术宽中胶囊调控转录因子蛋白家族核因子κB信号途径对功能性消化不良大鼠十二指肠微炎症的影响

摘　　要：目的:探讨枳术宽中胶囊对功能性消化不良(functional dyspepsia,FD)大鼠十二指肠微炎症的干预效应。方法:将36只SD大鼠随机分为空白组与造模组,通过复合造模法(碘乙酰胺灌胃+小平台站立)建立FD大鼠。将成功建模的FD大鼠随机分为模型组、莫沙必利组和低、中、高剂量枳术宽中胶囊组。给药2周后,观察各组大鼠一般状况、检测各组大鼠体质量及胃排空率,采用酶联免疫吸附试验法测定各组大鼠血清中IL-1β、IL-18的含量,以蛋白质印迹法检测NF-κB蛋白的表达水平。结果:与模型组比较,各剂量枳术宽中胶囊组与莫沙必利组大鼠经药物干预后毛发逐渐整洁光亮,自主活动频率增加,反应速度加快,粪便恢复正常,且体质量均呈上升状态(P<0.05)。苏木精-伊红染色显示,各组大鼠胃组织形态结构完整,细胞排列整齐。与模型组比较,各剂量枳术宽中胶囊组与莫沙必利组大鼠十二指肠苏木精-伊红染色肠绒毛排列整齐,炎症细胞浸润减少,细胞间隙较清晰。与模型组比较,中高剂量枳术宽中胶囊组与莫沙必利组大鼠血清IL-18及IL-1β含量明显降低(P<0.05),且中高剂量枳术宽中胶囊组与莫沙必利组大鼠十二指肠组织中的NF-κB蛋白表达亦明显降低(P<0.05)。结论:枳术宽中胶囊可以有效抑制十二指肠微炎症。Objective:To investigate the interventional effect of Zhizhu Kuanzhong capsule(ZZKZ)on duodenal microinflammation in rats with functional dyspepsia(FD).Methods:Thirty-six SD rats were randomly divided into the normal group and the model group,and the FD rats were established by the composite modeling method(iodoacetamide gavage and small platform standing).The successfully modeled FD rats were randomly divided into the model group,mosapride group and low,medium and high dose ZZKZ group.After 2 weeks of drug administration,the general condition of rats in each group was observed,the body mass and gastric emptying rate of rats in each group were detected.The levels of IL-1βand IL-18 in the serum of each group were determined using an enzyme-linked immunosorbent assay,and the expression levels of NF-κB proteins were detected by Western blotting.Results:Compared with the model group,rats in ZZKZ group and the mosapride group showed improvements after drug intervention.Their fur became gradually clean and shiny,spontaneous activity frequency increased,response speed quickened,stool returned to normal,and body weight showed an upward trend(P<0.05).Hematoxylin-eosin staining revealed that the gastric tissue structure of rats in all groups remained intact,with cells arranged neatly.Compared with the model group,rats in ZZKZ group and the mosapride group had duodenal villi arranged neatly,with reduced infiltration of inflammatory cells and clearer intercellular spaces in hematoxylin-eosin stained duodenal tissue.Compared with the model group,the serum levels of IL-18 and IL-1βin the medium and high dose ZZKZ group and mosapride group were reduced(P<0.05).The expression of NF-κB protein in duodenal tissues was reduced in both the medium and high dose ZZKZ group and the mosapride group compared with the model group(P<0.05).Conclusion:Zhizhu Kuanzhong capsule can effectively inhibit duodenal microinflammation.
