摘  要:骨肉瘤是儿童和青少年最常见的原发性恶性骨肿瘤,具有高度侵袭性,易在短时间内发生肺或全身转移,且复发率高,预后差。骨肉瘤的系统和规范治疗方案包括新辅助化疗、手术和术后放化疗。由于缺乏骨靶向性,传统的化疗药物往往不能在骨肉瘤部位积累,因此必须使用大剂量药物,进而不可避免地导致相关的全身不良反应。通过水凝胶载药系统进行局部给药,能够有效增高局部药物浓度、延长药物滞留时间、降低全身不良反应、减少给药频率、改善患者依从性。文章主要就近年来水凝胶载药系统在骨肉瘤治疗中的应用进行综述。Osteosarcoma is the most common primary malignant bone tumor in children and adolescents,characterized by high invasiveness,rapid progression to pulmonary or systemic metastasis within a short period,high recurrence rates and poor prognosis.The systematic and standardized treatment regimen for osteosarcoma includes neoadjuvant chemotherapy,surgery,and postoperative radiotherapy and chemotherapy.Due to the lack of bone targeting,traditional chemotherapy drugs often cannot accumulate at the site of osteosarcoma,necessitating the use of high-dose medications,which inevitably leads to related systemic adverse reactions.Local administration through hydrogel drug delivery systems can effectively increase local drug concentration,prolong drug retention time,reduce systemic adverse reactions,decrease frequency of administration,and improve patient compliance.This paper mainly reviews the application of hydrogel drug delivery systems in the treatment of osteosarcoma in recent years.
