摘  要:目的 探讨益生菌对肝硬化腹水大鼠肠道机械屏障和肝脏合成功能的影响。方法 40只雄性SD大鼠,随机选取30只大鼠建立肝硬化腹水模型,8周后30只造模大鼠随机分为模型组、多烯磷脂酰胆碱(PPC)组和益生菌组,每组10只;剩余10只大鼠为正常组,模拟造模过程。模型组和正常组给予饮用水灌胃,PPC组给予PPC混悬液灌胃,益生菌组给予双歧杆菌三联活菌混悬液灌胃,每日1次,连续4周。比较四组大鼠腹水量和肝脏指数。采用ELISA法检测四组血清转铁蛋白(TRF)及脂多糖(LPS)水平。免疫组织化学染色检测四组大肠组织紧密连接蛋白ZO-1的表达,计算ZO-1阳性指数。结果 模型组大鼠体重、肝脏重量、血清TRF水平及大肠组织ZO-1阳性指数均低于正常组,腹水量和血清LPS水平高于正常组(P<0.05)。益生菌组肝脏指数、血清TRF水平及大肠组织ZO-1阳性指数高于模型组,而腹水量和血清LPS水平低于模型组(P<0.05)。益生菌组体重低于PPC组,大肠组织ZO-1阳性指数高于PPC组(P<0.05)。结论 益生菌可能通过上调大肠组织ZO-1表达来增强肠道机械屏障,从而降低血清LPS水平,促进肝脏合成功能的修复。Objective To explore the effects of probiotics on the intestinal mechanical barrier and liver synthesis in cirrhotic ascites rats.Methods Of 40 male SD rats,30 male SD rats were randomly selected,in which a liver cirrhosis ascites model was established.After 8 weeks,30 modeling rats were randomly divided into three groups of A(given drinking water by gavage,10 cases),B(given polyene phosphatidylcholine suspension by gavage,10 cases)and C(given Bifidobacterium triple live bacterial suspension by gavage,10 cases).The group D of remaining 10 rats was simulated the modeling process and given drinking water by gavage.All intragastric treatments were performed once a day for 4 weeks.The abdominal water volume and liver index of four groups were compared.Serum levels of transferrin(TRF)and lipopolysaccharide(LPS)in four groups were detected by ELISA.The expression of tight junction protein ZO-1 in large intestine tissues of four groups was detected by immunohistochemical staining,and the positive index of zO-1 was calculated.Results Compared with group D,the body weight,liver weight,serum TRF levels and ZO-1 positive index of large intestine were lower,and the levels of abdominal water volume and serum LPS were higher in group A(P<0.05).The liver index,serum TRF level and ZO-1 positive index of large intestine in group C were higher than those in group A,while abdominal water volume and serum LPS levels were lower than those in group A(P<0.05).The body weight of group C was lower than that of group B,and the ZO-1 positive index of large intestine was higher than that of group B(P<0.05).Conclusion Probiotics may enhance the intestinal mechanical barrier by upregulating the expression of ZO-1 in the large intestine,thereby reducing the serum LPS level and promoting the repair of liver synthesis function.
